수중 휘발성 유기물질 분석을 위한 Headspace 법의 응용성에 관한 연구

dc.description.abstract[한글] 정수처리 과정에서 생성되거나 산업폐수의 하천유입 등으로 인해 발생하여 음용수중에 존재하는 휘발성 유기물질은 안전한 상수공급에 있어 새로운 관심의 대상이 되고있다. 휘 발성 유기물질에 대한 관심이 고조되면서 이를 분석해 내기 위한 분석방법들이 다양하게 개발되어 이용되고 있으며 현재 우리나라에서는 트리할로메탄류의 분석에 용매추출법과 h eadspace 법이 이용되고 있다. Headspace 법은 생체시료나 식품 등 다양한 시료 물질들로부터 휘발성 유기물질 분석에 이용되며 고체시료증의 수분함량 분석에도 이용된다. 또한 공기, 물, 토양, 슬러지 등과 같은 환경시료 중에 함유되어 있는 휘발성 유기오염물질 분석에도 응용되어 왔다. 이러 한 headspace 법은 purge and trap 법이라 불리는 dynamic headspace 법과 static headsp ace 법으로 분류되며 일반적으로 headspace 법이라 하면 static headspace 법을 말한다. 수중 휘발성 유기물질의 분석에 있어서 headspace 법은 시료성분이 기체상과 액체상 사이 에서 평형에 도달하게 된다는 사실에 기초하며 액체상과 기체상 사이의 화합물의 분포가 headspace 법을 이용한 수중 휘발성 유기물질 분석에 영향을 미치게 된다. 액체상과 기체 상 사이의 화합물의 분포는 온도, 각 화합물의 중기압, 수용해도, 분배계수 등에 영향을 받으며 용기내의 액체상 부피에 대한 기체상의 비에 영향을 받는다. 본 연구에서는 수중 휘발성 유기물질 분석을 위한 headspace 법의 영향요인들을 고려하 여 각요인의 headspace 분석에 미치는 영향을 조사하고 이들 요인들이 headspace 법의 분 석 검출한계에 어떠한 영향을 미치는 가를 조사 분석하였다. 수중 휘발성 유기물질 분석에 이용되는 headspace법의 분석감도 향상을 위해 분석감도 에 영향을 미치는 요인을 선정하여 분석한 결과 다음과 같은 결과를 얻었다. 1. 분석대상 항목의 시료부피 변화에 따른 분석감도 변화의 실험결과 상비 0.6인 10ml 시료에서는 5ml시료에 비해 1.30-2.84배의 감도가 증가하였으며 상비 0.7인 13ml시료의 경우는 1.26-3.21배의 감도가 증가하였다. 2. 온도조건 변화가 검출감도에 미치는 영향을 분석한 결과 온도가 증가함에 따라 피크 면적이 증가하였으며 40℃일때 1.10-1.54, 50℃일때 1.11-2.33, 60℃일때 1.12-3.28, 70 ℃일때 1.13-4.02배로 본 연구의 실험적 한계인 70℃까지 검출감도가 증가하였다. 3. 휘발성 유기물질 표준시료에 무기염(NaCl)을 첨가하여 검출감도에 대한 영향을 분석 한 결과 무기염 첨가량을 증가시킴에 따라 크로마토그래프 감응이 증가하였다. 포화점 첨 가량인 30-40% 첨가시까지 검출감도가 증가하였으며 그 이후 일정한 수준으로 유지되었다 . 4. 시료의 상평형 도달에 대한 흔들어줌의 영향을 분석한 결과 흔들어준 경우와 흔들어 줌 없이 정치시킨 경우간에 동일한 수준으로 검출되었다. 5. 조건변화를 통하여 headspace 법의 검출한계를 조사한 결과 일반적 실험조건인 경우 보다 영향요인을 변화시켜 실험한 경우가 실험적 한계값내에서 민감도가 높게 나타났다. 물질별로는 tetrachloroethylene을 제외한 대부분의 물질이 2배에서 2.5배 민감도를 나타 냈다. 이상의 결과로 보아 headspace법을 이용한 수중 휘발성 유기물질 분석시 온도조건, 시 료부피, 무기염 첨가 등을 통해 분석의 강도를 2배에서 4배까지 향상시킬 수 있는 것으로 평가되며, 본 연구의 실험적 한계내에서 검출한계는 각 물질 모두 2배 이상 향상시킬 수 있는 것으로 평가된다. [영문] Volatile organic compounds, produced in the water purification process or from industrial waste water, exist in the drinking water. This volatile organic compounds are now becoming our major concerns in the safe water supply. As the concerns for volatile organic compounds are growing, various analysis methods have been developed and used. For analyzing trihalomethanes, solvent extraction and headspace method are recommended to be used for field survey and monitoring in our country. Headspace method has been used for analysing volatile organic compounds in various sample matrix such as biological sample or foods and used for analyzing water content in solid sample, It has also been applied to analyze volatile organic compounds contained in environmental sample such as air, water, soil, and sludge. This headspace method is divided into dynamic headspace method which is called purge and trap method and static headspace method, but generally when we say headspace method it means static headspace method. In analysing volatile organic compounds in water, headspace method is based on the fact that the sample components can reach the equilibrium between liquid phase and vapour phase and the compounds distribution between these two phases affects the analysis results of the compounds. Compounds distribution between vapour phase and liquid phase is influenced by temperature, vapour pressure of each compounds, water-solubility and partition coefficient and also influenced by the ratio of vapour phase to liquid phase volume in the container. This study was conducted to examine the effect of each factors on headspace analytical sensitivity in consideration of influencing factors of headspace method for analysing volatile organic compounds in water. Also, effects of these factors on detection limit was examined. The results were obtained as follows ; 1. The 10 ml sample(phase ratio 0.6) shows 1.30-2.84 times higher sensitivity compared to 5 ml sample, and 13 ml sample(phase ratio 0.7) shows increase 1.26-3.21 times before. 2. The temperature went higher, the peak area increased. Detection sensitivity increased up to 70℃ which is the experimental limit in this study and 1.10-1.54 times higher at 40℃, 1.11-2.39 at 50℃, 1.12-3.28 at 60℃, 1.13-4.02 at 70℃ compared to 30℃ sample. 3. As increasing the quantity of NaCl, chromatographic resoponse increased. Detection sensitivity were increasing until reaching 30-40%(saturating point) and after that detection sensitivity were maintained a fixed level. 4. The result of detection reaches almost identical level whether the shaking case or shaking-less level. 5. In the case of general experimental condition, the detection limit was determinated at concentration from tetrachloroethylene 0.02 ppb to bromoform 0.4 ppb. In the case of considering influencing factors, the detection limit was determinated at concentration 0.016ppb(tetrachloroethylene, 1, 1, 1-trichloroethane, carbon tetrachloride) and bromoform 0.2 ppb. According to above results, it was estimated that the sensitivity in analysing can be improved two to Pour times by varying temperature condition, sample volume, adding NaCl in analyzing volatile organic compounds in water using headspace method. It was also estimated that within this study's experimental limit, detection limit can be improved core than two times in each substance case.-
