Abstract
Polyamine has been known to devote to cellular proliferation and growth in peripheral nerve regeneration and this study has been done to investigate the effect of polyamine and the interaction of nerve growth factor which is known as nerve cell growth promoting substance. Saphenous nerve of the rat has been injured with hemostat and polyamine, antinerve growth factor and DFMO knave been injected. After certain period, axon diameter and myelin thickness have been measured in proximal and distal portion of the saphenous nerve. 1. Axon diameter and myelin thickness decreased until 2 months begin to and increased 3 months after injury, however, they were not recovered to normal sizes even after 7 months. 2. In 1 month after polyamine treatment, proximal axon diameter increased and they were larger than normal group, however, 7 months group didn't show increment. Myelin thickness of polyamine group showed no difference with normal control group, while they were increased compared to experimental control group. In distal portion, axon diameter and myelin thickness were smaller than normal control, while they were larger than experimental control group. 3. In polyamine and anti-NGF group, axon diameters of the proximal portion were increased in the early period and after that, no changes has been shown. However, myelin thickness were decreased significantly and they were similar to experimental control group. In distal portion, axon diameter and myelin thickness were decreased and they were not recovered after certain period. 4. In DFMO group, axon diameter and myelin thickness of the proximal portion decreased in the early period and they were recovered after that. In 7 month group, they were almost same as normal control group. In the distal portion, axon diameter and myelin thickness were decreased and they were never recovered after 7 months. Axon diameters were increased in polyamine group and polyamlne-anti-NGF group both in proximal and distal group. It could be assumed that polyamine has the effect on the regeneration of both proximal and distal portion. Myelin thicknesses were increased in polyamine group, however, they were not changed in polyamine-anti-NGF group. With this results, it could be assumed that NGF has more effects on myelin thickness in regeneration. With the above results, it could be concluded that polyamlne has the effects in the early period of regeneration.
