췌장 선세포에서 cerulein에 의한 cytokine 발현의 신호전달체계

[한글]

친염증성 cytokine이 췌장염의 발생 및 임상경과에 관련되어 있지만 초기 단계의 조절기전에 대해서는 체계적인 연구가 아직까지 미미한 실정이다. 따라서 본 연구에서는 AR42J 세포주에 cerulein을 처리한 췌장염 세포모델에서 친염증성 cytokine인 IL-6, IL-1β,TNF-α의 발현이 ras, MAP kinase, NF-κB, AP-1 등에 의해 영향을 받고, 또한 ras와 MAP kinase가 NF-κB와 AP-1의 상위조절자임을 규명하고자 하였다. Ras, NF-κB, AP-1을 억제하기 위해 AR42J 세포에 Ras N-17, IκB mutant, TAM67을 transfection하였고, 대조군으로 야생종 세포와 pcDNA를 transfection한 세포를 이용하여 비교 분석하였다. 그리고 MAP kinase의 역할은 cerulein에 의한 ERK, JNK, p38 MAPK 활성과 ERK와 p38 MAPK에 대한 약리학적 억제제를 전처치한 후 NF-κB와 AP-1 활성 그리고 cytokine 발현을 측정하여 다음과 같은 결과를 얻었다.

1. AR42J 세포에 cerulein (10-8M)을 처치하였을 경우 cytokine (IL-6,IL-1β, TNF-α) mRNA는 cerulein 노출 1∼2시간 후에 발현되어 4시간까지 지속되었고, cytokine 단백질은 노출시간에 비례하여 발현이 증가하였다.

2. Ras N-17, IκB mutant, TAM67을 transfection하여 ras, NF-κB, AP-1을 억제하였을 때 cerulein에 의한 cytokine mRNA와 단백질 발현은 야생종 세포나 pcDNA를 transfection한 세포와 비교하여 현저하게 억제되었다.

3. Cerulein에 의한 NF-κB의 활성은 노출 30분내에 활성화되었고, 이후 감소하다가 4∼6시간 후에 다시 활성화되는 시간에 따른 2단계 활성을 보였고, Ras N-17, TAM67을 각각 transfection한 세포에서는 대조군 세포와 비교하여 cerulein에 의한 NF-κB 활성이 모두 억제되어 NF-κB 활성이 ras와 AP-1의 활성에 영향을 받았다.

4. Cerulein에 의한 AP-1의 활성은 노출 30분에 활성화되어 6시간동안 지속되었는데, Ras N-17를 transfection한 세포에서는 대조군 세포에 비해 cerulein에 의한 AP-1의 활성이 현저히 억제되었으나, IκB mutant를 transfection한 세포에서는 억제되지 않아 AP-1의 활성이 ras의 활성에는 영향을 받았으나 NF-κB 활성과는 무관하였다.

5. Cerulein 처치 후 3가지 형태의 MAP kinases (ERK, JNK, p38 MAPK)가 모두 조기에 활성화되었다.

6. U0126 또는 PD98059를 전처치하여 ERK를 억제하였을 때 cerulein에 의한 NF-κB와 AP-1활성 및 IL-6, IL-1β, TNF-α mRNA 발현은 전처치하지 않은 세포와 비교시 감소하였으나, SB203580를 전처치하여 p38 MAPK를 억제시에는 전사인자 활성 및 cytokine 발현이 감소하지 않았다.

이상의 결과로 AR42J 세포에서 cerulein에 의한 cytokine IL-6, IL-1β, TNF-α의 발현은 ras, MAPK, NF-κB, AP-1 등에 의해 조절됨을 알 수 있었다. 그리고 ras가 NF-κB와 AP-1경로의 상위 조절자이고, 또한 전사인자인 NF-κB와 AP-1는 MAP kinase 특히 ERK의 하위매개체일 것으로 추정된다. 또한 NF-κB의 활성에서 AP-1의 활성이 필요한 요소임을 규명하였다. 따라서 췌장 선세포에서 염증반응에 관여하는 cytokine 유전자 발현의 신호전달체계를 차단하는 치료가 개발된다면 췌장염의 경과를 완화시키는데 도움이 될 것으로 생각된다.





[영문]The signaling pathways mediating cytokine production in pancreatic acinar cells have not been fully understood. Recent studies indicated that cytokine expression requires activation of NF-κB and AP-1 as well as activation of the MAP kinases. However, the precise relationship between transcription factor and MAP kinase remains unclear. We examined the requirements of ras, MAP kinases, NF-κB, and AP-1 for cerulein-induced cytokine expresssion in pancreatic acinar AR42J cells. Cerulein was treated to the wild-type cells and the transfected cells with control vector (pcDNA), IκB mutant gene (IκB mt), H-ras mutant gene (Ras N-17), or c-jun dominant negative gene (TAM67). In addition, to investigate the role for MAP kinases, three subtypes of MAP kinases were measured in the cerulein-treated AR42J cells, and we used pharmacological inhibitors to attenuate signaling via these kinases. As a result, ① cerulein (10-8M) induced production of the inflammatory IL-6, IL-1β, and TNF-α mRNA and protein expression in AR42J cell. ② Inhibition of ras, NF-κB, and AP-1 using transfected cell with Ras N-17, IκB mutant, and TAM67 decreased the cytokine gene expression induced by cerulein (10-8M) as compared to pcDNA cells and the wild-type cells. ③ Cerulein induced NF-κB activation with biphasic kinetics. That is, NF-κB was strongly activated within 30 min after the stimulation and a second phase of NF-κB activation was prominent at 4∼6 h. Transfection of Ras N-17 or TAM67 in AR42J cells reduced cerulein-induced NF-κB activation. ④ Cerulein induced the activation of AP-1 within 30 min after stimulation and AP-1 activation was sustained continuously until 6 h. Transfection of Ras N-17 but not IκB mutant reduced cerulein-induced AP-1 activation. ⑤ Three subtypes of MAP kinases (ERK, JNK, and p38 MAPK) activities were elevated rapidly by cerulein in AR42J cells. ⑥ Inhibition of MEK activity resulted in a reduction of NF-κB and AP-1 activations, and cytokine expressions whereas the inhibition of p38 MAPK did not. In conclustion, cytokine gene expression by cerulein in AR42J cells was mediated via the activation of the ras, MAP kinases, NF-κB, and AP-1. Ras functioned as common upstream activators of both NF-κB and AP-1 pathway, and AP-1 might be required for activation of NF-κB. AP-1 and/or NF-κB transcription factors were potential downstream mediators of MAP kinases, especially ERK in the cerulein-induced cytokine expression in AR42J cell. It was believed that inhibition of signal transduction pathway such as ras, MAP kinases, NF-κB, and AP-1 might alleviate the inflammatory response in pancreatic acinar cells by suppressing cytokine gene expression.
