4-nitroquinoline-1-oxide(4NQO)유도 백서 구강암 발암과정에서 indomethacin이 langerhans세포와 T-임파구의 발현에 미치는 영향에 관한 연구

저자는 4-Nitroquinoline-1-oxide(4NQO) 유도 백서 구강암 발암과정에서 Indomethacin(IND) 투여시 면역기전에 관계되는 Lanserhans세포(LC)와 T -임파구의 분포를 관찰하기 위하여 생후 10주된 체중 150gm내외의 Sprague Dawley계 웅성백서 54마리를 정상군 6마리, IND을 투여한 대조군 12마리,4NQO를 주 3회 구개점막에 도포한 발암군 18마리, INO 투여와 4NQO도포를 병행한 실험군 15마리로 나누어, 정상군은 실험즉시와 11,21주째, 대조군은 3,5,7,9,11,21주째, 발암군과 실험군은 7주째부터 23주째까지 2주간격으로 각기 2마리씩 회생시켜 적출한 구개점막과 설점막을 ATPase 염잭표본, 광학현미경표본, 전자현미경표본및 T-임파구의 염색표본으로 제작 관찰한 바 다음과 같은 결과를 얻었다.

1. 광학현미경 적으로 4NQO만 도포한 발암군에서는 실험초기부터 상피의 중식과 rate peg의 증식이 시작되었고 7주째부터는 부전각흑상과 개개세포의 각화상 및 미약한 상피 이형성상과 임파구 침윤상을 보였다. IND 투여와 4NQO 도포를 병행한 실험군에서도 4NQO만 도포한 발암군에서보다 반응은 미약하나 상피층의 증식, 부전각화상 및 rete peg의 증식상을 보이며 임파구의 침윤상이 현저하였고 IND만 투여한 대조군에서는 정상군과 유사하였다.

2. 정상군에서는 실험기간에 따른 LC의 숫적 변화가 거의 얼었으나 IND만 투여한 대조군에서는3주째 현저한 LC의 증가상이 관찰되어 IND 투여와 LC의 분포화는 밀접한 관계를 보였다.

3. 4NQO만 도포한 발암군과 4NQO와 IND 투여를 병행한 실험군에서는 LC의 수가 현저히 감소된 소견을 보였으며 두군 공히 21주까지 계속 감소하였으나 실험군이 다소 완만한 감소를 보여 IND이 발암현상을 지연시키고 특히 종양면역에 관여함을 유추케 한다.

4. T-helper cell과 T-suppressor cell의 출현은 정상군과 대조군에서는 경미하고 불규칙한 상을 보였으며 임파구의 침윤과 비례하는 상을 보였다.

5. 발암군과 실험군 공히 T - helper cell의 출현이 T- suppressor cell의 출현보다 현저히 증가하였으나 실험군이 발암군에 비해 다소 경미하였다. 각군 공히 21주째에 가장 심한 침윤상을 보였으나 23주째는 감소현상을 보였고 전체적으로 불규칙한 출현상을 보였다.

This study was undertaken to investigate the effect of indomethacin on the distribution of Langerhans cells and T- Iymphocytes related with immune response of 4- Nitroquinoline-1-oxide induced carcinogenesis at the palate and tongue of albino

rat.

54 Sprague-Dawley stain 10 weeks old albino rats, about 150gm weighted, divided into a normal group of 6 rats without treatment, a control group of 12 rats given indomethacin, a carcinogenesis group of 13 whose palatal mucosa were applied with

4-Nitroquinoline -1-oxide three times a week and experimental group of 18 rats were treated with indomethacin and whose Palatal mucosa were applied 4- Nitroquinoline -1- oxide.

All these 54 rats were subjected to be observed as being ATPase stained specimens specimens for the observation of light and electron microscope, and T- Iymphocyte stained specimens.

The obtained results were summarized as fellows:

1. In carcinogenesis group, proliferation of epithelial layer and rete peg were observed early period of the experiment and skowed parakeratosis individual cell keratinization acanthosis and lymphocyte infi1tration from 13th week of the experiment on lightmicroscopically, while experimental group showed less reaction than that of carcinagensis group.

2. The number of Langeuhans cells in normal group rarely chanced until 21st week of the experiment while the Langerhans cells increased markedly from 3rd week of the experiment in control group.

3. The number of Langerhans cells were deceased markedly and persistantly until 21st week of the experiment both in us and experimental groups. However the number of Lanserhans cell in experimental group showed less decreasing pattern than that

of carcinogenesis group.

4. Appearance of the T-helper cells and T-suppressor cells were minimal and irregullar number both in normal and control group.

Thus it is assumed that administration of indomethacin and distribution of Langerhans cell showed close relation.

5. In carcinogenesis and experimental groups, the number of the T- helper cells was apparently increased than that of the T-suppressor cells, but increasing pattern in experimental group was lefts than in carcinogenesis group.

The cells increased most in the 21st week, decreased from the 23rd week and the appearance of these cells were irregular in general throughout the experiment.
