자가 적혈구의 Epstein-Barr Virus 감염에 의한 B임파구 세포 전환에 미치는 영향에 관한 연구

세포융합 방법을 이용하여 단클론항체 ( monoclonal antibody )생산을 할 수 있는 기술이 소개 된 이후 현재도 많은 학자들에 의해 이 방법이 이용되고 있다. 그러나 세포융합법에 의해 생산된 단클론항체는 주로 생쥐를 이용하기 때문에 인간에게 적용하기란 여러 가지 난점이 있다.

인간 B임파구를 선택적으로 감염시키는 Epstein- Barr virus(EBV)를 이용하여 인간 B임파구를 감염시켜 세포 전환을 유도하여 영구히 성장 증식되는 B임파구에 의하여 인간 특이 단클론항체를 생산함으로써 이러한 난점을 해결할 수 있을 것으로 본다.

그러나 EBV 감염에 의한 B임파구의 세포 전환과정에서 T임파구의 세포독성이 세포 전환된 B임파구를 인지하여 사멸시키기 때문에 세포주로의 확립에 어려움이 있다.

이에 본 논문에서는 인간 특이 단클론항체 생산을 위한 기초적인 연구로서 인간의 자가 적혈구를 첨가하였을 때 EBV 감염에 의한 B임파구 세포 전환에 미치는 영향 및 EBV 감염에 의한 B임파구 세포 전환에 있어 복강삼출세포 ( Peritoneal exudate cell ) 의 feeder layer effect를 조사하고자 실험을 하여 다음과 같은 결과를 얻었다.

1. 자가 적혈구와 함께 EBV를 분획한 B임파구에 감염시킨 결과 B임파구는 18.3 ± 4.9 × 10**4 으로 증식하여 대조군 14.0±2.9 × 10**4 에 비해 세포수는 증가되었으나 의의있는 차이는 없었다.

2. 적혈구의 농도가 EBV 감염에 의한 B임파구 세포 전환에 미치는 영향을 조사하고자 B임파구에 1%, 5% 및 10%의 적혈구와 함께 EBV를 감염시킨 바, B임파구 세포수는 각각 12.5 ± 2.1 x 10**3, 18.0 ± 2.9 × 10**3 및 14.5 ±0.7 × 10**3 으로 대조군 11.0± 1.4 × 10**3 에 비해 증식되었으나 각 군 사이의 의의있는 차이는 없었다.

3. 분획하지 않은 임파구용액을 E8V로 감염시켜 배양한 후 B임파구의 증식을 관찰한 바 B임파구는 3.3 ±0.8 × 10**4 으로 원래의 세포수 3 × 10**4 과 큰 차이가 없었으나 적혈구와 함께 감염시킨 군 7.0± 1.0 × 10**4 과는 의의있는 차이가 있었으며, 이에 비해 B임파구를 분획하여 감염시킨 대조군은 14.0±2.9 × 10**4 로 실험군에 비해 약 4배 증가된 반응도를 나타내어 의의있는 차이를 나타냈었다. 이와 같이 자가 적혈구를 이용함으로써 T임파구에 의한 B임파구 세포 전환 역행 현상을 억제할 수 있었으나 T임파구의 세포수를 말초혈액의 5배로 증가시켜 실험한 군에서는 적혈구 존재에 관계없이 모두 1 × 10**4 이하로서 원래의 세포수 보다 감소하였다.

4. 복강삼출세포의 feeder layer effect를 알아보고자 1 × 10**4 의 BALB/C 마우스 복강삼출세포를 사용한 바, 분획한 B임파구를 EBV단독으로 감염시킨 군, 적혈구와 같이 감염시킨 군 및 분획하지 않은 임파구를 적혈구와 같이 감염시킨 군 각각에서 12.3 ±3.4 × 10**4, 17.3 ±5.2 × 10**4 및 7.0± 1.0 × 10**4 과 큰 차이가 없었다.

위의 실험 결과에서 볼 때 EBV감염에 의한 B임파구의 세포 전환에 있어서 분획한 B임파구를 EBV로 감염시키는 방법보다 적혈구와 함께 감염시키거나 또는 전혈을 직접 감염시키는 것이 B임파구의 세포주 확립에 보다 나은 효율을 제공할 수 있다고 본다.

[영문]

After introduction of monoclonal antibody production by cell fusion, it has been used by many investigators. However, since antibodies produced by fusing of B lymphocytes and myeloma cells are originated from mice, it is foreign protein to human. Therefore, it is difficult to introduce the antibodies produced by the system for human body for clinical, especially therapeutic tools.

To resolve this problem, human specific monoclonal antibody would be the one of the best solutions. One of these methods is establishment of B lymphoblastoid cell line by use of Epstein-Barr virus (EBV) which selectively infects human B lymphocytes and immortalize them.

In spite of the advantageous fact, establishment of B lymphoblastoid cell line and its practical use remained under experimental state, mainly due to its low efficiency in establishment, because the T lymphocytes in the preparation recognize the membrane change of B lymphocytes induced by EBV and kill the transformed B lymphocytes.

Thus, the possible role of autologous erythrocytes, and peritoneal exudate cells in the process of transformation of B lymphocytes induced by EBV was studied to enhance the efficiency of the transformation of B lymphocytes induced by EBV.

The results summarized as follows:

1. EBV induced B lymphocytes transforamtion appeared to be enhanced by adding autologous RBC, even though significance was not satisfactory.

2. To determine the appropriate RBC concentration of maximal enhanced effect on B lymphocytes transformation, 1%, 5% and 10% RBC suspensions were applied. It was most effective at 5%, but significance was not satisfactory.

3. T lymphocytes inhibited B lymphocytes transformation induced by EBV, but the regression effect of T lymphocytes against B lymphocytes transformation was inhibited by autologous RBC. 5 folds of peripheral blood T lymphocytes abolished the RBC effect.

4. The feeder layer effect of BALB/C mouse peritoneal exudate cells was not observed.

It could be concluded that EBV induced B lymphocyte transformation in the presence of autologous RBC or whole blood by infecting with EBV directly may be of a great help in improving the establishment of B lymphoblastoid cell lines for the production of human specific monoclonal antibodies.
