Abstract

Human papillomavirus(HPV) type 16과 type 18은 자궁경부암 및 진행성 경향이 놓은 자궁경부 상피내 종양에서 주로 숙주세포 염색체 내에 삽입된 형태로 높은 출현빈도를 보이고 있으며, 이들 virus genome 중에서 특히 trans anting factoe 또는 cia-acting factor(promoter/enhancer)로 작용하는 부위의 세포변형 능력에 관한 연구가 최근 대단히 활발하다.

본 연구에서는 HPV type 16 genome 중 서포 형질변환능력을 갖는 부위라고 알려지는 E6, E7 open reading frame(ORF) 부위가 자궁경부 상피내 종양 또는 자궁경부암 조직 내어서 표현되는지의 여부를 in situ hybridination기법을 통해 알아보고자 하었다. 숙주세포 내에 삽입된 HPV type 16 DNA의 E6, E7 ORF로부터 전사되는 mRNA에 대해 특이적으로 결합할 수 있는 single stranded RNA probe를 제조하기 위해 E6, E7 ORF와 noncoding region 일부를 포함하는 1.326kb의 subgenomic fragment를 pGEM 4z 전사 vector의 mutiple cloning region(MCR) 내로 방향성을 가지게하여 subcloning하고, MCR양쪽에 위치한 SP6또는 T7 promoter에 특이적으로 작용하여 RNA를 전사하는 SP6 또는 T7 RNA polymerase를 이용하여 시험관내에서 RNA probe릅 합성하었다. in situ hybridization을 시행한 결과 자궁경부암으로 진단된 환자의 생검조직 내상피층에서 target mRNA-biotinylated RNA probe-Extravidin alkaline phosphtase conjugate 복합체와 nitro blue tetrazolium(NBT), 5-bromo-4-chloro-3-indolyl phosphate(BCIP)가 반응하여 생기는 signal이 미만적으로 세포질 내에 분포하는 양상을 보였으나, 기질(stroma)내에는 전혀 signal이 발견되지 않았다. 상피세포 전층은 모두 비정형세포로 대치되어 있었고, 표층 상피세초에서는 공동세포(koilocyte)도 관찰 되었다. Signal 의 강도는 중상층 상피세포 부위에서 가장 강하게 나타나는 것을 괄찰할 수 있었다.

이상의 결과로 저자는 HPV type 16 DNA의 E6, E7 ORF 부위가 자궁경부암 조직 내에서 국소적으로 활발히 전사되는 것을 확인하였다.

Human papillonavirus(HPV) type 10 and type 18 DNAs are known to be found frequently in the biopsy specimens from the lesion Of both Cervical Carcinoma in situ (CIS) with high tendency for progression and invasive cervical carcinoma(ICC), and their integration into human genome hus been documented. It has been

suggested that some regions within the viral genome may play some role as a trans-acting factor or a cia-acting promoter/enhancer, and efforts are now directed to show the transforming activity of these regions. Also it was reported that HPV type 16 E7 ORF encodes a protein of transcriptional transactivation and cellular

transformation function analogous to those of adenovirus EIA proteins. The purpose of this study is to detect the RNA transcripts transcribed from the HPV type 16 E6, E7 ORF regiong in CIN or ICC via in situ hybridization technique, To make single

stranded RNA probe which specifically hybridize to mRNA from E6, E7 ORF of HPV type16 DNA inserted into host cell chromosomel 1.326kb subgenomic fragment was subcloned into multiple cloning region of pGEM 4z transcription vector with orientation and using SP6 or T7 promoter specific RNA polymerasel RNA probe was synthesized in vitro. After in site hybridization, epithelia of tissue biopsy section from patient diagnosed as carcinoma in situ revealed dark blue signal resulted from the reaction between target mRNA-biotinylated RNA probe-Extravidin alkaline phosphatase conjugate and chromogenic substrate nitro blue

tetrazolium(NBT) , 5-bromo-4-chloro-3-indolyl phosphate(BCIP). Signals were distributed diffusely in entire epithelium but were not detected in stroma. Entire epithelia was replaced by dysplastic cells, and there were some koilocytes in superficial epithelia. Signal intensity was most strongly observed at upper third of epithelia.

This study demonstrated that HPV type 16 E6, E7 ORF activelyt ranscribed in carcinoma of the cervix.
