대장암 세포주에서 비스테로이드성 항염증 약물에 의한 caspase-3의 활성화

Abstract
[한글]

역학적인 조사를 통하여 비스테로이드성 항염증 약물(nonsteroidal anti-inflammatory drug, 이하 NSAIDs로 약함)이 대장암의 발생빈도와 사망률을 감소시키고, 가족성 대장 용종증 환자에서 용종의 크기와 수를 감소시킨다고 보고되고 있다. NSAIDs가 대장암의 발생과 진행을 억제하는 기전이 세포증식을 억제하거나 세포고사(apoptosis)를 유발하기 때문이라는 주장이 대두되고 있으나 NSAIDs가 어떤 기전으로 세포고사를 유발하는지에 대해서는 아직 알려진 바가 거의 없다. 본 연구에서는 NSAIDs에 의한 대장암 세포고사의 분자 기전을 규명하기 위하여 각종 세포에서 여러 자극에 의한 세포고사의 실행과정에서 중요하다고 알려진 caspase-3가 활성화되는지 알아보았다.

세포고사는 DAPI (4', 6'-diamidine-2'-phenylindole dihydrochloride) 염색 후 confocal microscopy로 관찰한 세포의 형태와 agarose gel 전기영동에서 DNA fragmentation으로 확인하였으며, cell free system을 이용하여 고사세포에서 추출한 세포질 단백질이 정상적인 핵의 고사를 일으킬 수 있는지 알아보았다. Western blot으로 caspase-3와 그 기질인 poly (ADP-ribose) polymerase (PARP)의 분해를 알아보았으며, CASP-1 (ICE), CASP-2(hIch-1) 및 CASP-3 (caspase-3) 등의 CASP 유전자 mRNA는 RT-PCR과 RNase protection assay로 측정하였다. Caspase-3 활성도는 Ac-DEVD-CHO (acetyl-Asp-Glu-Val-Asp-aldehyde)를 이용하여 억제하였으며, fluorogenic peptide substrate인 Ac-DEVD-AMC를 이용하여 caspse-3 활성도를 측정하였다.

Indomethacin 및 sulindac으로 처치한 HT-29 세포에서 추출한 세포질이 Hamster 간에서 분리한 정상적인 핵의 DNA fragmentation을 유도하는 것으로 보아 NSAIDs에 의하여 고사가 유발된 세포의 세포질내에서 어떤 물질이 활성화되어 세포고사의 특징적인 핵의 변화를 일으킨다는 사실을 확인하였다. Fluorogenic peptide substrate인 Ac-DEVD-AMC를 이용하여 측정한 caspse-3 활성도는 투여한 NSAIDs의 농도와 시간에 비례하여 증가하였다. HT-29의 indomethacin 매개 세포고사에서 caspase-3가 중요한 역할을 한다는 것은 투여한 indomethacin의 용량과 시간에 비례한 caspase-3의 감소와 caspase-3의 기질인 PARP의 단백분해를 Westem blot으로 측정함으로써 확인하였다. 또한 caspase-3 억제제인 Ac-DEVD-CHO는 HT-29에서 indomethacin에 의한 DNA fragmentation을 억제하였다. 그러나 indomethacin이 CASP-3를 포함한 CASP 유전자의 mRNA 발현을 증가시키지는 않는 것으로 보아 indomethacin 매개 세포고사에서 caspase-3의 활성화는 CASP-3 유전자의 전사와 이에 따른 단백질합성의 증가에 기인하는 것은 아니며 이 효소가 전사 및 합성된 후 단백질 구조의 변화에 의하여 활성화됨을 알 수 있었다.

[영문]

Epidemiological studies have demonstrated that nonsteroidal anti-inflammatory drugs(NSAIDs) decrease the incidence of and mortality from colon cancer. In addition, NSAIDs reduce the number and size of polyps in patients with familial adenomatous polyposis. The mechanisms responsible for the anti-neoplastic effect of NSAIDs are yet far from complete understanding but one of possible mechanisms is an induction of apoptosis. In this study, we have explored the role of caspase-3, a major apoptosis executing enzyme, in NSAIDs-induced apoptosis of colon cancer cell line, HT-29. In a cell free system, cytosolic protein extracted from indomethacin-treated HT-29 cells induced DNA fragmentation of normal nuclei, isolated from Hamster liver, indicating that NSAIDS activate certain cytosolic component which can induce nuclear changes specific for apoptosis. In addition,

indomethacin treatment in HT-29 cells induced a dramatic increase in caspase-3-like protease activity measured by a cleavage of fluorogenic substrate, Ac-DEVD-AMC. Dose- and time-dependent decrease of caspase-3 as well as cleavage of its substrate poly (ADP-ribose) polymerase (PARP) further demonstrated indomethacin-induced activation of caspase-3. Indomethacin-induced DNA fragmentation was dose-dependently attenuated by caspase-3-like protease inhibitor, Ac-DEVD-CHO.

However, mRNA expression of CASP genes was not influenced by the addition of indomethacin highlighting the importance of post-trans-rational modification of this enzyme for the activation. These results suggest that NSAIDs including indomethacin induce apoptosis in colon cancer cells through a caspase-3-dependent

mechanism, which may contribute to the chemopreventive functions of these agents.
