자외선 조사가 Listeria monocytogenes의 감염에 대한 감수성과 싸이토카인 생성에 미치는 영향

[한글]

자외선 조사가 면역기전에 미치는 영향으로는 세포성 면역반응을 억제하는 것으로 보고되고 있으나 그 작용기전에 대해서는 명확히 밝혀져 있지 않다. 최근 싸이토카인(cytokine)에 관한 연구가 활발하여 짐에 따라 면역반응이 싸이토카인에 의해 조절되며 특히 세포성 면역반응은 주로 림프구 및 대식세포 각각에서 생성되는 interferon(IFN)-γ 및 interleukin(IL)-12에 의해 유도된다는 사실이 밝혀져 있다.

이에 본 연구자는 자외선 조사가 세포성 면역반응에 미치는 효과와 그 작용기전을 규명하고자 하였다. 세포성 면역반응이 비교적 높은 것으로 알려져 있는 C57B/6 마우스에 자외선 B를 조사하고 세포조직내 기생세균인 Listeria monocytogenes로 접종하여 숙주의 저항성 변화를 관찰하였다. 다른 한편으로는 자외선 B로 조사한 마우스 림프구의 IFN-γ 생성 및 대식세포의 tumor necrosis factor(TNF)-α 및 IL-12의 생성 변화를 조사하여 다음과 같은 결과를 얻었다.

1. 자외선 B 조사 3일후 마우스 비장지수를 측정하였는데, 자외선 비조사군의 비장지수 1.000에 비교하여 7.5 KJ/m**2의 자외선 조사군에서 비장지수가 1.585로서 59%증가하였고, 15 KJ/m**2의 자외선 조사군에서 비장지수가 1.959로서 자외선 비조사군에 비하여 96% 증가하였다.

2. 자외선 B 7.5 KJ/m**2 조사 2일후 Listeria monocytogenes를 접종하고, 비장을 적출하여 균수를 세어 본 결과 접종 2일후 자외선 조사군이 4.5×10**5/ml, 비조사군이 1.7×10**5/ml로서 자외선 조사군이 비조사군에 비해 2.6배 증가하였고, 접종 3일 후에는 자외

선 조사군이 6.8x10**5/ml, 비조사군이 1.5x10**5/ml로서 자외선 조사군이 비조사군에 비해 4.5배 증가하였다.

3. IFN-γ의 생성은 자외선 B 7.5 KJ/m**2 조사 3일후 마우스 비장세포를 concanavalin A(Con A)로 자극시 자외선 조사군이 4480.0±0.3 ng으로 비조사군의 5934.0±0.2 ng에 비해 24% 감소하였고 Listeria로 자극시 자외선 조사군이 1061.0±0.6 ng으로 비조사군의 2108.0±0.5 ng에 비해 50% 감소하였으나, 자외선 B 7.5 KJ/m**2 조사 7일후 Con A 자극시 자외선 조사군이 6086.8±0.4 ng 비조사군이 6080.6±0.3 ng이며 Listeria로 자극시 자외선 조사군이 5328.7±0.2 ng 비조사군이 5652.5±0,5 ng으로 7일 후에는 자외선 조사에 의한 차이가 없었다.

4. TNF-α의 생성은 자외선 B 7.5 KJ/m**2 조사 3일후 마우스 복강 대식세포를 lipopolysaccharide(LPS)로 자극시 자외선 조사군이 73.3±0.5 ng으로 비조사군의 105.0±0.4 ng에 비해 30% 감소하였고 Listeria로 자극시 자외선 조사군이 43.5±1.0 ng으로 비조사군

의 94.0±0.2 ng에 비해 54% 감소하였다. 자외선 B 7.5 KJ/m**2 조사 7일 후 LPS로 자극시 자외선 조사군이 86.8±0.5 ng 비조사군이 102.2±0.6 ng으로 15% 감소되었으나 Listeria로 자극시 자외선 조사군이 41.0±0.2 ng으로 비조사군 92.0±7.7 ng에 비해 55% 감소

하였다.

5. IL-12의 생성은 자외선 B 1.5 KJ/m**2조사 3일후 마우스 복강 대식세포에서 Con A로 자극시 자외선 조사군이 7.7±0.4 ng으로 비조사군의 15.4±0.3 ng에 비해 50% 감소하였으며, Listeria로 자극시 자외선 조사군이 11.6±0.2 ng으로 비조사군의 18.1±0.2 ng에

비해 36% 감소하였으나 자외선 B 7.5 KJ/m**2 조사 7일후 Con A로 자극시 자외선 조사군이 15.4±0.4 ng 비조사군 14.6±0.2 ng이며 Listeria로 자극시 자외선 조사군이 14.2±0.3 ng 비조사군이 13.1±0.4 ng으로 7일 후에는 자외선 조사에 의한 차이가 없었다.

이상의 결과로 보아 자외선 B 조사후 마우스 비장 지수 증가 및 Listeri monocytogenes의 감염에 대한 감수성 증가는 IFN-γ, TNF-α 및 IL-12의 생성 저하와 관련이 있는 것으로 생각되며, 이러한 싸이토카인의 생성 저하는 세포성 면역기전의 저하를 일으키는 것으로 사료된다.

Although the effect of ultraviolet (UV) irradiation on immune response was reported to supress cellular immune response, the exact mechanism was not elucidated. As recent development in cytokine research progress, it is well-known that immune response is regulated by cytokines and especially cellular immune response is induced by interferon (IFN)-γ and interleukin (IL)-12 which is mainly produced from lymphocytes and macrophages respectively.

Therefore our purpose was to elucidate the UV effect on cellular immune response and its mechanism. We have investigated the changes of host resistance by injection of Listeria monocytogenes which is an intracellular parasite after UVB irradiation in C57BL/6 mice which is kown to have relatively strong cellular immune response.

In addition we also have investigated the changes in the production of IFN-γ from lymphocytes and the production of tumor necrosis factor (TNF)-α and IL-12 from macrophages in mice by UVB irradiation. The results are as follows.

1. The mouse spleen index(non-irradiated group 1.000; 7.5 KJ/m**2 UVB irradiated group 1.585; 15 KJ/m**2 UVB irradiated group 1.959) was calculated 3 days after UVB irradiation. The increase was 59% by 7.5 KJ/m**2 UVB irradiation and 96% by

15KJ/m**2 UVB irradiation.

2. The number of Listeria monocytogenes in spleen increased 2.5 times 2 days after inoculation(irradiated group 4.5×10**5/m1; non-irradiated group 1.7×10**5/ml) and 4.5 times 3 days after inoculation (irradiated group 6.8×10**5/ml; non-irradiated group 1.5×10**5/m1), which was irradiated by 7.5 KJ/m**2 UVB 2 days before inoculation, compared with the non-irradiated control

group.

3. IFN-γ production in mouse spleen cells 3 days after 7.5 KJ/m**2 UVB irradiation by concanavalin A(Con A) stimulation (4480.0 ±0.3 ng) decreased 24% compared with the non-irradiated control group (5934.0±0.2 ng) and the production after irradiation by stimulation with Listeria(1061.0±0.6 ng) decreased 50% compared with the non-irradiated control group (2108.0±0.5 ng). IFN-γ production in mouse spleen cells 7 days after 7.5 KJ/m**2 UVB irradiation by Con A stimulation (6086.8±0.4 ng) was not significantly different from the non-irradiated control group (6080.6±0.3 ng) and the production after irradiation by stimulation with Listeria (5328.7±0.2 ng) was not significantly from the non-irradiated control of group (5652.5±0.5 ng).

4. TNF-α production in mouse peritoneal macrophages 3 days after 7.5 KJ/m**2 UVB irradiation by lipopolysaccharide (LPS) stimulation (73.3±0.5 ng) decreased 30% compared with the non-irradiated control group (105.0±0.4 ng) and the production

after irradiation by stimulation with Listeria(43.5±1.0 ng) decreased 54% compared with the non-irradiated control group (94.0±0.2 ng). TNF-α production 7 days after 7.5 KJ/m**2 UVB irradiation by stimulation with LPS (86.8±0.5 ng) decreased

15% compared with the non-irradiated control group (102.2±0.6 ng) and the production after irradiation by stimulation with Listeria(41.0±0.2 ng) decreased 55% compared with the non-irradiated control group(92.0±0.7 ng).

5. IL-12 production in mouse peritoneal macrophages 3 days after 7.5 KJ/m**2 UV irradiation by Con A stimulation(7.7±0.4 ng) decreased 50% compared with the non-irradiated control group(15.4±0.3 ng) and the production after irradiation by

stimulation with Listeria(11.6±0.2 ng) decreased 36% compared with the non-irradiated control group (18.1±0.2 ng). IL-12 production in mouse peritoneal macrophages 7 days after 7.5 KJ/m**2 UVB irradiation by Con A stimulation (15.4±0.4 ng) was not significantly different from the non-irradiated control

group(14.6±0.2 ng) and the production after irradiation by stimulation with Listeria(14.2 ±0.3 ng) was not significantly different from the non-irradiated control group(13.1±0.4 ng).

These results suggest that the increase of mouse spleen index and susceptibility of Listeria monocytogenes infection was correlated with the decreased production of IFN-γ, TNF-α and IL-12, which was known to induce the suppression of cellular

immune response.
