채소류에 대한 발암성 농약 잔류와 그 건강 위해성 평가

현대의 인간은 의학의 발달과 생활수준의 향상으로 일반적인 건강 수준 및 수명은 크게 신장되었으나, 부적절한 생활양식 등에서 기인하는 암 발생률 또한 증가 일로에 있으며 특히, 암 사망요인의 35% 이상이 식품섭취와 관련되어 있는 것으로 추정되고 있다.

우리나라는 60년대 이후 급격한 산업화와 더불어 환경오염이 가속화되고 사회적 관심과 국민의 우려가 커짐에 따라 이러한 오염물질의 과학적인 위해성 평가방법과 기준설정 및 관리대책에 대한 연구가 진행되어 오고 있다. 또한 현실의 인간은 각종 복합 유해화합물에 다중 노출되므로 종합적인 환경오염관리를 위하여 각 노출경로를 포괄하는 연구를 필요로 하지만, 기존의 연구들은 환경오염 경로중 주로 대기 및 수질을 중심으로 진행되어 온 관계로 경구를 통한, 즉 오염된 식품섭취를 대상으로 하는 위해성 평가는 상대적으로 미진한 실정이다.

따라서, 식품 중에서 양적 비중이 상대적으로 크며 생식의 빈도가 높아서, 생산과정중 사용된 농약 오염으로 인한 인체 위해성의 가능성이 많은 채소류를 대상으로 10종을 선정하여 각각 15건 총 150건의 시료를 시장에서 무작위 표본추출하고, 유력한 인체 발암성

농약 10종에 대한 시료분석을 통하여 검출된 4종을 중심으로 안전 한계지표의 비교와 위해성 평가를 실시하였다.

20종의 농약에 대한 위험성 확인에서 유력한 인체 발암물질(B)은 aldrin, T-BHC, chlorbenzilate, chlordane, DDT, dieldrin, folpet, heptachlor, heptachlor epoxide 및 toxaphene, 가능한 인체 발암물질(C)로는 parathion, Not classifiable(D)은 endrin, 검토중

인 물질(Under review)로는 alachlor, captan, chlorothalonil, dicofol, daminozide, endosulfan, lindane 및 mirex를 분류하였다.

환경 노출평가는 DDT, dieldrin, folpet 및 heptachlor epoxide가 9건(6%)에서 0.0006 - 0.09ppm(평균 잔류량; 0.00001 ~ 0.0007) 검출되었으며, 특히 heptachlor epoxide는 마늘의 국내 및 FAO/WHO의 잔류농약 허용기준을 각각 초과하였으나 평균 잔류량은 모두 허용기준 미만이었다. 또한 일일 추정 섭취량은 0.0009 ~ 0.079㎍으로서, DDT가 가장 많이 섭취될 수 있다.

인체 노출평가는 발생 가능한 모든 노출량을 산정하기 위해서 scenario를 가정하였는데, scenario Ⅰ은 불검출 시료에 대하여 각 농약의 검출 한계값을 적용하여 가장 보수적인 가정을 하였으며, scenario Ⅱ는 불검출 시료에 대하여 검출 한계값의 1/2을 적용하였고

또한 scenario Ⅲ은 검출된 오염도만을 적용한 경우이다. 인체 노출량은 scenario I(2.1 * 10**-6 ~ 2.2 * 10**-5 mg/kg/day)에서 가장 높았으며, scenario Ⅱ(1.1 * 10**-6 - 1.2 * 10**-6)의 약 2/3 ~ 2배 및 scenario Ⅲ(1.3 * 10**-8 ~ 1.3 * 10**-6)의 약 3.5 ~ 50배로서 DDT, heptachlor epoxide, folpet 및 dieldrin의 크기 순서였다.

IRIS의 발암력과 HSDB의 ADI를 이용한 용량-반응 평가에서 발암력은 3.5 * 10**-3 ~ 1.6 * 10(mg/kg/day)**-1 로서 dieldrin, heptachlor epoxide, DDT 및 folpet의 순서로 크게 나타났다.

ADI는 0.0005 - 0.01(mg/kg/day)로서 folpet, DDT, dieldrin 및 heptachlor epoxide의 순으로 높았으며, 실제 안전용량은 1.2 * 10**-3 - 1.1로서 folpet, DDT, heptachlor epoxide 및 dieldrin의 크기 순으로 추정되었다.

각 scenario에 따라서 산출한 초과 발암 위해도 범위는 risk I에서 1.1 * 10**-8 - 5.5 * 10**-5로 가장 높았으며, rick Ⅱ는 7.0 * 10**-9 ~ 3.2 * 10**-5 그리고 risk Ⅲ은 3.1 * 10**-9 - 8.8 * 10**-6 이었다. 농약별 risk는 heptachlor epoxide(8.8 * 10**-6 ~ 5.5 * 10**-5 ), dieldrin (2.1 * 10**-7 ~ 3.4 * 10**-5 ), DDT(4.4 * 10**-7 ~ 7.5 *10**-6 ) 그리고 folpet (3.1 * 10**-9 ~ 1.1 * 10**-8 )의 크기 순으로 산출되었다. Heptachlor epoxide는 실제 채소류 섭취에 의한 평균 노출량만을 고려한 경우에도 10**-6 을 초과하고 있으나, folpet은 세가지 scenario에서 모두 아주 낮은 위해도를 나타냈다. DDT와 dieldrin은 risk Ⅲ의 경우 10**-6 을 초과하지는 않지만 비교적 높았으며, risk Ⅰ과 risk Ⅱ에서는 4.1 * 10**-6 ~ 3.4 * 10**-5 이었다.

EDI/ADI는 가장 낮은 folpet 0.009%로부터 가장 높은 heptachlor epoxide 0.2%의 비율로서 매우 낮은 수준이다.

한국 MRL을 적용한 경우, TMDI/ADI는 heptachlor epoxide 0.1%로부터 dieldrin 0.38%였으며, EDI/TMDl는 folpet 0.03%로부터 heptachlor epoxide 1.88%였다. 또한, FAO/WHO의 MRL을 적용시 TMDI/ADl는 folpet 0.19%로부터 dieldrin 3.5%였고, EDI/TMDI는 dieldrin 0.004%로부터 heptachlor epoxide 0.48%의 수준이었다. 그러므로 TMDI/ADl에서 Folpet만이 0.26%로서 FAO/WHO의 0.19%보다 약간 높은 안전 한계지표를 보였으며, EDi/TMDl는 dieldrin과 heptachlor epoxide가 FAO/WHO의 수준 보다 약 4배가 높게 나타났다.

이상의 결과에서, heptachlor epoxide는 채소류 이용시 감소계수 자료의 축적과 더욱 현실성 있는 식이 섭취량의 추정 등의 섭취량 경감을 위한 조치를, 그리고 DDT와 dieldrin은 체계적인 monitoring이 필요할 것으로 생각된다. 따라서, 우선 각 유해물질 및 식품군별로 체계적인 위해성 평가를 실시하여 안전마진 확보에 필요한 과학적 근거를 축적하고 그 결과를 토대로 국민의 건강과 사회적 손익을 신중히 검토한 후에 국제기준의 수용여부를 결정할 것을 제안한다.

[영문]

Risk assessment and safety margin index(SMI) were evaluated on carcinogenic pesticides(CPs) in 150 specimens from ten kinds of vegetables which randomly sampled at Karak agricultural products market in Seoul.

In the hazard identification for 20 pesticides: aldrin, T-BHC, chlorbenzilate, chlordane, DDT, dieldrin, folpet, heptachlor, heptachlor epoxide and toxaphene were classified as probable human carcinogen(B).

Parathion was possible human carcinogen(C), while endrin was Not classifiable(D). As the Under review: alachlor, caftan, chlorothalonil, dicofol, daminozide, endosulfan, lindane and mirex were selected.

As the result of environmental exposure assessment: DDT, dieldrin, folpet and heptachlor epoxide were detected in 9 samples(6%) in the range of 0.0006 ~ 0.09ppm(average residues:0.00001 ~ 0.0007).

Particularly, heptachlor epoxide was exceeded maximum residue limits(MRLs) of pesticide both Korea and FAO/WHO, however all the average residues were within the limits. The most ingested pesticide was DDT among the estimated daily intake(EDI, 0.0009 - 0.079㎕).

Three scenarios were hypothesized for human exposure assessment. Scenario I was the most conservative hypothesis which supposed the undetected CPs is detection limit values, scenario ll was assumed the undetected CPs as half of the detection limit values, and scenario 111merely applied the detected values of CPs.

Human exposure value of scenario I(2.1 * 10**-6 ~ 2.2 * 10**-5 mg/kg/day) was the greatest as 2/3 ~ 2 times of scenario Ⅱ(1.1 * 10**-6 ~ 1.2 * 10**-5 ) ant 3.5 ~ 50 times of Scenario Ⅲ(1.3 * 10**-8 ~ 1.3 * 10**-6 ). The order of exposure magnitude was DDT, heptachlor epoxide, folpet, and dieldrin.

The carcinogenic potency was 3.5 * 10**-3 ~ 1.6 * 10(mg/kg/day)**-1 and the order of magnitude was dieldrin, heptachlor epoxide, DOT, and folpet. Acceptable daily intake(ADI) was 0.0005 ~ 0.01(mg/kg/day) and the magnitude was folpet, DDT, dieldrin, and heptachlor epoxide as the order. The order of virtually safe dose(VSD, 1.2 * 10**-3 ~ 1.1ppm) magnitude was folpet, DDT, heptachlor epoxide, and dieldrin.

Excess cancer risk(ECR) which calculated from the exposure scenarios was 1.1 * 10**-8 ~ 5.5 * 10**-5 in risk Ⅰ, 7.0 * 10**-9 ~ 3.2 10**-5 in risk Ⅱ, and 3.1 * 10**-9 ~ 8.8 * 10**-6 in risk Ⅲ. The order of ECR magnitude in CPs was heptachlor epoxide(8.8 * 10**-6 ~ 5.5 * 10**-5 ), dieldrin (2.1 * 10**-7 ~ 3.4 * 10**-5 ), DDT(4.4 * 10**-7 ~ 7.5 * 10**-6 ), and folpet (3.1 * 10**-9 ~ 1.1 * 10**-8 ). Heptachlor epoxide was exceeded 10**-6 , while folpet was very low in all the three risk. DDT and dieldrin were within 10**-6 in the risk Ⅲ, but risk Ⅰ and risk Ⅱ were relatively high level(4.1 * 10**-6 ~ 3.4 * 10**-5 ).

EDI/ADl was very low level as the range of 0.009%(Folpet) ~ 0.2%(heptachlor epoxide).

Based on the Korean MRL, theoretical maximum daily intake(TMDI)/ADl was 0.1%(heptachlor epoxide) ~ 0.38%(dieldrin) and EDI/TMDI was 0.03%(folpet) ~ 1.88%(heptachlor epoxide). In case of the FAO/WHO MRL, TMDI/ADI was 0.19%(folpet) ~ 3.5%(dieldrin) and EDl/TMDl was 0.004%(dieldrin) ~ 0.48%(heptachlor epoxide). Therefore, only the folpet(0.26%) in TMDI/ADl was slightly exceeded FAO/WHO(0.19%). Dieldrin and heptachlor epoxide in EDI/TMDl were four times higher than FAO/WHO.

According to the results, we'd better take measures for reducing the heptachlor epoxide intake by the data accumulation on reduction Factor and practical estimate of CPs in vegetables. Furthermore, DDT and dieldrin should be monitored continuously.

Consequently, we must systematically evaluate the cost/benefit on national interests and health promotion by the scientific bases of safety margin which derived from the risk assessment of each pesticides and food group. Thereafter, we can decide whether accept the FAO/WHO MRLs in Korea.
