백서에서 혈청 prolactin 변화조작이 성선 및 부성선에 미치는 영향

Abstract
[한글] 웅성 백서에서 prolactin과 성선 및 부성선과의 형태학적이고 기능적인 관계를 추구하고자, 성년기의 정상백서 25마리를 대상으로 고proloactin 혈증과 저prolactin 혈증을 유발시키기 위하여 자성 혹은 웅성의 4개 뇌하수체를 웅성의 신피막하에 동종이식하고, bro mocriptine(2mg/day)을 투여하였다. 조작한지 80일후 고환정맥내와 하대정맥내의 혈청 testosterone치의 변화를 hCG투여전과 투여후에 Solid Phase-RIA로 관찰하였고, Leydig 세포 수와 세정관의 단면적, 전립선세포의 장경을 전산화 형태계측으로 측정하였다. 뇌하수체 이식군에서 대조군과 bromocriptine 투여군보다 혈청 testosterone이 감소하였고(p〈0.0005, p〈0.005), hCG 10 I.U를 4시간 간격으로 2차례 피하주사 후 40시간에 측정한 혈청 testosterone치도 감소하였으며, 대조군과의 비교는 통계적 의의가 없었다(p〈0.1, p〈0.01). 또 세정관의 단면적은 차이가 없었으며 Leydig 세포수는 감소하였고(p〈0.0005, p〈0.0005), 전립선 세포의 장경도 감소하였다(p〈0.0005, p〈0.0005). Bromocriptine 투여군은 대조군보다 혈청 testosterone치가 증가하였고(p〈0.0005), hCG 투여후 혈청 testosterone치도 증가하였다(p〈0.05). 세정관의 단면적은 차이가 없었으며, Leydig 세포수는 증가하였고(p〈0.0005), 전립선세포의 장경도 증가하였다(p〈0.005). 뇌하수체 이식군과 bromocriptine 투여군에서 각각의 대조군 사이에 각 계측에서 통계적 의의가 있는 차이는 볼 수 없었다. 이상의 결과는 고prolactin 혈증이 성선 steroid의 음성 되먹 임기전에 대한 시상하부-뇌하수체 축의 반응을 증가시키고, bromocriptine이 성선에서의 steroid생성에 간접 혹은 직접 영향을 미친다는 것을 시사하고 있다. In contrast low plasma levels of prolactin induced by treatment with bromocriptine in adult male rat resulted in increased plasma levels of testosterone, increased number of Leydig cells and increased height of prostatic glandular cells. The area of the seminiferous tubule was not changed under any circumstance. And the basal plasma levels of testosterone in testicular vein were proportioned to its levels in I.V.C. It suggests that adrenal gland has minimal effects on action of prolactin to gonad. The change of basal plasma level of testosterone accompanied by the change of numbers of Leydig cells under induced hyperprolactinemia and hypoprolactinemia suggest that prolactin affects the testis including the anatomical change. These results support that hyperprolactinemia appear to sensitize the hypothalamic-pituritary axis to the negative feedback effects of gonadal steroids and that bromocriptine has indirect or direct effects on steroidogenesis of testis.
