Parathion, Chloroform 및 2, 6-Dichlorophenol의 중독에 의한 흰쥐의 혈액학적 소견과 면양적혈구에 대한 항체 생산세포수에 미치는 영향

Abstract
[한글] 환경오염은 금세기에 해결하여야만 하는 심각한 과제중의 하나이다. 그중 농약에 있어 parathion 그리고 상수원수의 오염에 따른 정시처리시 염소소독에 의해 생성되는 chloroform chplorophenol 등은 인체에 독작용을 미치는 매우 유독한 화학물질이므로 더욱 중요 한 문제로 대두되었다. Parathion, chloroform과 2,6-dichlorophenol의 중독작용은 생체 대사기능 등을 억제 또는 감퇴시킨다고 하며 이는 직접 또는 간접적으로 질병의 병발에 관여한다는 것을 의미한다. 따라서 저자는 parathion, chloroform, 2,6-dichlorophenol의 중독이 면역능력에 미치는 영향을 연구 조사하였다. 실험동물소는 건강한 흰쥐를 사용하였으며 독성물질로는 흰쥐의 무게 kg당 parathion 1.3㎎/olivedb 10㎖, chloroform 100㎎/olivedb 10㎖을 복강내투여하였고 또한 2,6-dichlorophenol 13㎎/olivedb 10㎖은 경구투여하였다. 각 독성물질 투여후 3주 경과후에 동량을 동법으로 재투여하였고 면양적혈구를 항원으로 2일간격 3회 투여하여 면역시킨후 4주때에 흰쥐에서 적출한 비장세포부유액의 항체생산능력을 보체에 의한 면양적혈구 용혈반응의 정도로 측정하였다. 이상의 실험결과 다음과 같은결론을 얻었다. 1. 백혈구수의 변화 ; Parathion, chloroform, 2,6-dichlorophenol을 투여한 실험군과 이를 투여하여 종독시 킨후 면양적혈구로 면역시킨 실험군에서 대체로 대조군과 실험군사이에 있어 혈액중 백혈구수에는 유의한 차이가 없었다. 2. Hemoglobin 량의 변화 ; Parathion, chloroform을 투여하여 1주 경과후 실험한 군에서 대조군에 비해 hemoglobin 량이 감소하였고 이독성물질들을 3주때 재투여하고 면양적혈구로 면역시켜 4주때 실험한 군에서도 hemoglobin 량이 감소하였다. 그러나 2,6-dichlorophenol을 투여하고 1주 경 과후 실험한 군에서는 대조군과 유의한 차이가 없었으며 이독성 물질을 3주때 재투여하고 면양적혈구로 면역시켜 4주때 실험한 군에서는 대조군에 비해 hemoglobin 량이 감소하였다. 3. Hamaticrit의 변화 ; Parathion, chloroform을 투여하여 1주 경과후 실험한 군에서 대조군에 비해 hematocrit가 감소하였고, 또한 이독성물질들을 3주때 재투여하고 그후 면양적혈구로 면역시켜 4주때 실험한 군에서도 hematocrit가 감소하였다. 그러나 2,6-dichlorophenol을 투여하여 1 주 경과후 실험한 군에서는 대조군과 유의한 차이가 없었으나 이독성물질을 3주때 재투여하고 면양적혈구로 면역시켜 4주때 실험한 군에서는 대조군에 비해 hematocrit가 감소하였다. 4. 혈청단백중 총단백, albumin, α-globulin, β-globulin, γ-globulin, A-G(Albumin/Globulin)의 변화 ; Parathion, chloroform, 2,6-dichlorophenol을 투여하여 1주, 2주 경과후 실험한 결과 대조군과 유의한 차이가 없었다. 5. 용혈플라크수의 변화 ; Parathion, chloroform, 2,6-dichlorophenol을 투여하고 3주때 면양적혈구로 면역시켜 4주때 실험한 군에서 대조군에 비해 면역반응 용혈플라크수의 생성이 적었다. 이상의 실험결과를 보아 parathion, chloroform 및 2,6-dichlorophenol에 의해 중독되면 적혈구 생성수가 감소되며 면역능력에 감퇴를 가져 올 것으로 사료된다. A Study on Immune Response by Intoxication of Parathion, Chloroform and 2,6-Dichlorophenol in Rats. Among the environmental pollutants, parathion, chloroform and 2,6-dichlorophenol may impair human health ; they may inhiboit or reduce the metabolic function of human body and may furthermore cause diseases derectly or indirectly. And this study was undertaken to investigate the effects on the immune response by intoxiocation of parathion, chloroform and 2,6-dichlorophenol. Parathion (1.3 ㎎/1㎏, olive oil 10㎖), chloroform(100㎎/㎏, olive oil 10㎖) were administered via intraperitoneal injection to rats. And 2,6-dichlorophenol (13㎎/㎏, olive oil 10㎖) was administered via oral injection. After 3 weeks, the rats were intoxicated with the above chemicals and immunized with RBC. After 4 weeks, the immune response of rat spleen cells was measured by the Jerne's technique. The results were obtained as follows ; 1. There was no change of leukocyte counts by the intoxication of parathion, chloroform and 2,6-dichlorophenol. 2. Parathion, chloroform and 2,6-dichlorophenol reduced hemoglobin contents for most intoxicated an immunized groups. 3. Hematocrits were decreased by the intoxication of parathion, chloroform and 2,6-dichlorophenol significantly. 4. It was determined that total protein, A/G (Albumin/Globulin), α-, β- and γ-globulins in rat serum were not changed. 5. Intoxication by paratasion, chloroform and 2,6-dichorophenol reduced the number of hemolytic plaque to the sheep RBC in rat spleen cells. Therefore, the capacity of erythrocyte production and the immune response of rat spleen cells were decreased by the intoxication of parathion, chloroform or 2, 6-dich-lorophenol.
