DMBA 도포가 HAMSTER 협낭상피의 LANGERHANS세포에 미치는 영향에 관한 실험적 연구

Abstract

[영문] [한글] 저자는 DMBA유도 조직 변화시 Langerhans 세포의 분포변화를 관찰하여, 종양조직에서의 Langerhans 세포의 면역학적 기능을 연구하려는 목적하에, 생후 6주된 Golden Hamster 3 0마리를 대조군 12마리와 실험군 18마리로 나누어, 대조군에는 heavy mineral oil을, 실 험군에는 heavy mineral oil에 녹인 0.5% 9, 10 Dimethyl-1, 2-Benzanthracene(DM-BA)을 주3회 협낭에 도포한 후, 6, 8, 10, 12, 14, 16주에 대조군은 2마리, 실험군은 3마리씩 각각 희생시켜 채취한 표본에 H-E 및 PAP염색을 시행한 후 검경하여 다음과 같은 결론을 얻었다. 1. H-E염색시 대조군에서는 정상조직과 별 차 없었으며, 실험군에서는 기간이 지남에 따라 점막의 과각화층, 단핵염증세포 침윤, 유두종, 이형성 및 상피내암 소견이 관찰되었 다. 2. PAP염색시 대조군의 점막 및 점막하조직과 실험군의 점막에서는 Langerhans 세포를 발견할 수 없었다. 3. 실험군 6주째인 이형성 발현시 Langerhans 세포가 이형성부위 주위에 나타나 8주째 급격히 증가하다가 이후 상피내암 발현전까지 다소 감소한 상을 보였다. 4. 실험군에서 이형성이 진행되는 동안 평균 5내지 17개 정도의 Langerhans 세포가 점 막하조직 혈관주위에서 관찰되었다. 5. 실험군 14주째인 상피내암 발현시 Langerhans 세포가 상피내암 주위보다는 떨어진 정상 점막하조직에 더 많이 보였다가 16주째는 급격히 감소하여 상피내암 주위에서는 관 찰할 수 없었다. THE EFFECT OF DISTRIBUTION OF LANGERHANS CELLS IN DMBA-PAINTED HAMSTER CHEEK POUCH EPITHELIUM YU JI HO Department of Dental Science, Graduate School, Yonsei University. (Directed by Prof. Eui Wung Wung, D.D.S., M.S., Ph.D.) The purpose of this study was to investigate the distribution of Langerhans cells on Hamster buccal pouch painted with DMBA (9, 10 Dimethyl-1, 2-benzanthracene). Golden Hamsters-6 week old-were used in this experimental study and devided into control and experimental groups. Experimental groups were painted with 0.5% DMBA in heavy mineral oil on right buccal pouch three times weekly. The animals were sacrificed at every 6, 8, 10, 12, 14, 16th week and excised buccal mucosa for microscopic examination. All specimen stained with Hematoxylin-Eosin and PAP method, and examined by light microscope. The obtained results from this study were as follows. 1. In H-E stain, hyperkeratosis, papillomatous growth, dysplasia, carcinoma in situ, and inflammatory cells were observed in experimental groups. 2. In PAP stain, Langerhans cells were not observed under the submucosa and mucosa of control group and the mucosa of experimental groups. 3. At the early stage of dysplasia in 6th week, Langerhans cells were observed around the dysplasia and markedly increased in 8th week, but slighty decreased in 10 and 12th week. 4. During the formation of dysplasia, Langerhans cells were observed under the submucosa(average no. 5-10), especially aggregated around the blood vessels in cellular dysplastic area. 5. After the formation of carcinoma in situ, Langerhans cells were observed more frequently under the normal submucosa than the peripheral area of cancer tissue and it showed markedly decreasing tendency under the normal submucosa in 16th week and was not observed around the carcinoma in situ.
