골유합과정에서 골아세포의 대사와 증식은 골형성과 밀접한 관계가 있다고 생각되고 있다. 특히 골아세포내 Ca**2+ 활성도의 증가가 골형성 작용에 관여하는 것으로 알려져 있다. 세포내 Ca**2+ 농도는 세포막에 존재하는 Ca**2+ 이온 통로를 통한 유입 외에도 Ca** 2+ ATPase 활성도, Na**+ -Ca**2+ 교환이동기전 및 Na**+ -K**+ ATPase 활성도에 의해 영향을 받을 수 있다. 이 중 Na**+ -K**+ ATPase 활성도에 의해 조절되는 세포내 Na**+ 농도는 Na**+ -Ca**2+ 교환이동기전을 통해 세포내 Ca**2+ 농도를 조절한다.
본 연구의 목적은 골아세포막의 Na**+ -K**+ ATPase 활성도를 측정하여 그 특성을 규명하고, ouabain이 골아세포막 Na**+ -K**+ ATPase 활성도와 골아세포의 분화에 미치는 영향을 관찰하는 것이다. 이에 쥐의 경골에서 분리하여 배양된 골아세포의 세포막에서 Na**
+ -K**+ ATPase 활성도를 측정하고, Na**+ -K**+ ATPase 활성도를 억제시키는 것으로 알려진 ouabain에 의한 Na**+ -K**+ ATPase 활성도의 변화를 관찰하고, 한편으로는 횐 쥐의 경골 골절 부위에 ouabain 처치 골아세포를 주입하여 골절 유합 과정을 시기별로 관찰하
4. 골절 후 7일째 골절조직의 Hematoxylin-eosin 염색상 ouabain 처치군은 대조군보다 골아세포 및 유골조직 형성이 더 풍부한 양상을 보였다.
5. 항 bromodeoxyuridine(BrdU) 마우스 항체를 이용한 면역조직화학적 염색결과 대조군에서는 골절 10일째 BrdU 표지지수가 높았으며, ouabain 처치군에서는 골절 7일째에 높고 이후에는 감소하였다.
이와같은 연구결과로 보아 골아세포막 Na**+ -K**+ ATPase 활성도를 감소시키는 ouabain은 골절 초기에 골아세포의 분화를 촉진시켜 골형성을 증가시키는 것으로 사료된다.

Proliferation and Metabolism of osteoblasts play an important role in osteogenesis for bone healing. An increase in intracellular Ca**2+ activity of osteoblast cell, is known to modulate the activity of osteogenesis. Intracellular Ca**2+ activity can be controlled directly by Ca**2+ influx through Ca**2+ channel and Ca**2+ efflux via Ca**2+ ATPase, and also indirectly by Ca**2+ -K**+ ATPase which generates Na**+ gradients for Na**+ -Ca**2+ exchange. Thus, in this study, to investigate the relations between Na**+ -K**+ ATPase in osteoblastic membrane and osteogenesis, the effects of ouabain on Na**+ -K**+ ATPase activity in osteoblast were measured along histologic changes of bone healing by H-E and BrdU stain. The results obtained in this study are as follows:
1. The measured Vmax of Na**+ -K**+ ATPase in osteoblastic membrane was 59.1±26.4nmole Pi/mg protein/hr, and Km was 4.6±3.5 mM.
2. Increasing concentrations of ouabain from 10**-8 M to 10**-5 M decreased the activity of Na**+ -K**+ ATPase, with significant inhibition at 10**-5 M.
3. Increasing concentrations of vanadate from 10**-8 M to 10**-5 M decreased the activity of Na**+ -K**+ ATPase, with significant inhibition at 10**-6 M and 10**-5 M.
4. H-E stain showed more abundant osteoblast and making the maturation of woven bone faster in the ouabain-treated than in the control group especially on 7th day after fractire.
5. BrdU labeling index showed more active osteoblastic proliferation in the ouabain-treated than in the control group especially on 7th day after fractire.
In conclusion, ouabain inhibited the Na+-K+ ATPase activities of osteoblast and enhanced the osteogenesis in early period after fracture.
