Abstract
[한글]척추동물의 골격근 섬유는 형태학적으로 분류하기도 하였으나 Needham(1926)과 Pater 등(1972)은 근육 내 myoglobin의 함량에 따라 red, white 그리고 intermediate형으로 분류하였고 Close(1967)는 근 수축속도에 따라 slow와 fast형으로 분류하였으며 Engel(1962)은 myosin ATPase의 조직화학적 염색법으로 type Ⅰ(slow 근 섬유에 해당)과 type Ⅱ(fast 근 섬유에 해당)로 분류하였다.

ATPase의 조직화학적 염색법은 Padykula와 Herman(1955a)에 의하여 처음 고안되었으며 그 후 몇가지 형태로 발전되어 왔다. 대표적인 것으로 Guth와 Samaha(1969, 1970)의 방법이 있는데 이것은 type Ⅰ의 myosin ATPase가 산성에 안정하고 alkali에는 불안정하며 type Ⅱ의 myosin ATPase는 type Ⅰ과 반대되는 특성을 갖고 있음을 이용하여 myosin ATPase의 조직화학적 염색 전에 근육박편을 acid 또는 alkali 용액에서 preincubation시킴으로써 type Ⅰ 또는 type Ⅱ의 myosin ATPase 중 하나를 선택적으로 불활성화시켜서 양자의 구별을 뚜렷하게 하는 방법이다. 그러나 이 때 중요하다고 알려진 Preincubation 용액의 pH와 preincubation 시간을 발표된 방법과 동일하게 조절하여도 그 염색 반응이 일정하게 나타나지 않는 수가 많다.

따라서 저자는 preincubation시 온도 변화가 myosin ATPase 염색반응에 어떻게 영향을 미치는 지에 대하여 연구하였다.

본 실험에서는 성숙된 백서(150∼250g)를 사용하였다. 백서를 두부 강타하여 희생시킨 즉시 족저근과 가재미 근을 분리하여 acetone과 고체 이산화탄소의 혼합액 속에 넣어 급격히 동결시켰다. 각 근육을 적당한 크기(5㎜ × 5㎜)로 잘라서 mounting chuck 올려놓고

0.C.T. compound로 매몰시킨 후 -20℃로 조절된 cryostat 내에 보관하였다가 10μ의 두께로 cut하여 받침유리 위에 올려놓고 실온에서 30∼240분간 건조시켰다.

Acid preincubation시에는 건조된 근육표본을 여러가지 pH(pH 4.0에서 4.8까지 0.1 단위의 차이를 두었음)의 acid preincubation 용액에 넣고 온도(21℃, 25℃, 28℃, 34℃)와 시간(각 온도에 따라 그 시간 범위가 다르지만 전체적으로 20초에서 90분까지)을 달리하면서 preincubation하였다.

Alkali preincubation시에는 건조된 근육표본을 먼저 3∼5℃ buffered formaldehyde 용액(5% formaldehyde, 200mM sodium cacodylate, 68mM CaCl^^2, 340mM sucrose, pH 7.6)에 넣어서 5분간 고정하였다. 이 고정된 근육표본을 세척액(0.iM tris-HCl, 18 mM CaCl^^2, pH 7.8)에 넣고 잘 흔들어 주면서 1분간 씻어낸 다음 근육표본에 묻어 있는 용액을 압지로 제거하고 이를 여러가지 pH(pH 8.8에서 10.3까지 0.3 단위의 차이를 두었음)의 alkali preincubation 용액에 넣고 온도(20℃, 25℃, 30℃, 35℃)와 시간(각 온도에 따라 그 시간 범위가 다르지만 전체적으로 30초에서 150분까지)을 달리하면서 preincubation하였다.

Alkali preincubation 용액의 조성은 0.025 M borax와 18mM CaCl^^2였는데 0.1N HCl 또는 0.1N NaOH를 가하여 원하는 pH로 조정되었다. Preincubation이 끝난 다음 근육표본을 세척액(0.1M tris-HCl, 18mM CaCl^^2, pH7.8)에 넣고 잘 흔들어 주면서 1분씩 두 번 씻어내고 조직표본에 묻어 있는 용액을 압지로 제거한 후 후술하는 바와 같은 요령으로 incubation 하였다.

Incubation은 34∼37℃에서 Gomori medium (18mM CaCl^^2 20mM sodium barbiturate, 5mM ATP, pH9.2∼9.4)에 30분 동안 하였으며 다음 과정인 염색은 Guth와 Samaha(1970)의 방법대로 시행하였다.

위와 같은 방법으로 근육표본을 염색하여 다음의 결과를 얻었다.

1. Alkali preincubation을 위하여 formaldehrde 용액으로 근육을 고정할 때 고정액의 온도를 3∼5℃로 하면 그 처리시간은 5분이 적당했지만 20℃로 상승시키면 15초가 적당했고 2분이 지나면 myosin ATPase의 활성화가 나타나지 않았다.

2. Acid preincubation의 경우 온도를 일정하게 할 때 pH가 낮아질수록(pH 4.0∼4.8 범위에서 pH 4.0에 가까울수록) 염색반응에 필요한 preincubation 시간이 짧아졌다.

3. Alkali preincubation의 경우 온도를 일정하게 할 때 pH가 높아질수록(pH 8.8∼10.3 범위에서 pH 10.3에 가까울수록) 염색반응에 필요한 preincubation 시간이 짧아졌다.

4. Acid preincubation의 경우나 alkali preincubation의 경우 모두에서 pH를 일정하게 할 때 온도가 상승될수록 myosin ATPase 염색반응에 필요한 preincubation 시간이 짧아졌으며, 이는 단순히 용액의 (OH**-)/(H**+)비가 온도에 따라 변하는 것에 기인된 것이

아니라 myosin ATPase 활성화와 preincubation 온도가 직접적인 관련이 있는 것으로 나타났다.



[영문]Vertebrate skeletal muscle fibers have been classified by several different methods. For instance, Needham(1926) and Peter et al.(1972) classified the muscle fibers into the red, white and intermediate types depending on the myoglobin content, Close(1967) classified them into the slow and the fast muscle fibers according to the speed of contraction, and Engel(1962) classified them into type Ⅰ(slow fiber) and type Ⅱ(fast fiber) on the basis of histochemical staining characteristics of the myosin ATPase.

Of various histochemical staining methods of the myosin ATPase, the method of Guth and Samaha(1969) is most widely used in the distinction between fiber typeⅠand Ⅱ. This method involves the preincubation of the tissue section in acid or alkali solution before staining, so that selectively inactivates the myosin ATPase in the type Ⅰ or type Ⅱ fiber. Although this procedure provides an useful tool In the fiber classification, the final results often vary depending on the condition of preincubation, such as pH. temperature and pre-incubation tome. We therefore

undertook In the present study to investigate the interrelationship between the pH, temperature and the time of preincubation and to establish conditions which assure

consistent results at wide varying experimental temperatures.

Albino rats, weighing 15O-250 g, were killed by a head blow and then the plantaris and soleus muscles were excised and immediately frozen in a bath of acetone with soled carbon dioxide. The belly of each muscle was ablated, plated on a mounting chuck with 0.C.T. compound and then kept in a cryostat(-20℃). Sections were cut at 10μ thickness, mounted on glass slides and allowed to dry in the air for 30-240 minutes. In acid preincubation, the muscle section was preincubated in 0.1M sodium acetate buffer(pH 4.0-4.8) at 21-34℃ for 20 seconds to 90 minutes, taken out and then blotted. In alkali preincubation the muscle section was first fixed in 5% formaldehyde buffer solution(5% formaldehyde, 200 mM sodium cacodylate, 68 mM CaCl**2, 340 mM sucrose) at 3-5℃ for 5 minutes. The fixative was removed by a 1 minute rinse in 0.1 M tris-HCl containning 18 mM CaCl**2(pH 7.8) and the excess solution drained on blotting paper. The alkali preincubation was carried out for 30 seconds to 150 minutes using 0.025 M borax(pH 8.8-10.3, titrated with 0.1 N HCl or NaOH) containing 18 mM CaCl**2 at 20-35℃.

After preincubation in acid or alkali solution, the muscle section was incubated in Gomori medium (18 mM CaCl**2, 20 mM sodium barbiturate, 5 mM ATP, pH 9.2-9.4) for 30 minutes at 34-37℃. And then the muscle section was stained for myosin ATPase.

The results are summarized as followings

1. In alkali preincubation, myosin ATPase activity was well demonstrated at 3-5℃ for 5 minutes or at 20℃ for 15 seconds when flexed tilth buffered formaldehyde solution.

2. In acid preincubation, the optimal preincubation time range for the histochemical staining of myosin ATPase became shorter as the pH(pH range, 4.0-4.8) decreased at any given temperature.

3. In alkali preincubation, the optimal preincubation time range for the histochemical staining of myosin ATPase became shorter as the pH(pH range, 8.8-10.3) increased at a given temperature.

4. When the pH of preincubation solution was fixed at a constant value, the optimal preincubation time range for the histochemical staining of myosin ATPase became shorter as the preincubation temperature increased.

These results indicate that preincubation temperature as well as the pH and the preincubation tome directly affect the histochemical staining response of myosin ATPase.
