Abstract
[한글]산업의 발달과 국민 소득 수준의 향상으로 생활에 필요한 화학물질이 양적으로나 질적으로 증가하고 있는 추세이다. 또한 인체 및 환경 생태계에 유해한 화학물질도 증가하는 추세에 있다. 선진 각국에서는 식품첨가물, 치료 약품 및 농약에 대한 안전성이 대두되기

시작하면서 이들에 대한 안전수준(일일 허용섭취량)율 규명하고, 음용수, 대기, 토양 및 산업장과 같은 환경매체내에서 검출되는 유독화학물질의 허용노출수준(환경기준)을 설정하여, 인체 및 생태계의 피해효과를 최소화하기 위한 방법론으로서 위해성 평가(risk assessment)가 실시되기 시작하였다.

그러나 우리나라의 경우는 기존 화학물질과 더불어 신종 화학물질의 증가에도 불구하고 화학물질에 대한 안전성 평가를 위한 관리체계가 미흡한 상태에 있다. 또한 화학물질이 증가함에 따라 환경매체내에는 유독화학물질이 검출되고 있으나 이러한 오염물질을 제어하기 위한 기준도 없는 상태이며, 있다고 할지라도 과학적이고 합리적인 근거에 의해 설정된 것이 아니기 때문에 타당성이 결여되어 있는 실정이다.

따라서 본 연구에서는 기존 또는 신종 화학물질의 안전성 평가제도로서, 환경오염물질의 기준설정 근거로서 건강 위해성 평가의 방법론을 구축하고 이 방법론(4가지 주요단계: 위험성 확인, 노출평가, 용량-반응 평가, 위해도 결정)에 입각하여, 현재 우리나라의 대기중에서 검출되고 있는 미량 오염물질의 대표적 물질이고 다환 방향족 탄화수소류의 지표물질인 benzo(a)pyrene(이하, BaP이라 칭함)에 대한 위해성 평가를 실시하여 다음과 같은 결론을 얻었다.

1. 위험성 확인: BaP는 동물에 대하여 발암성물질이다. 반면 사람에서의 발암성에 대한 증거는 다수의 역학 조사에서 BaP에 노출된 경우 증가된 비교위험도를 나타내었으나 흡연 및 BaP이외의 다른 발암성 물질에의 동시 노출과 같은 혼란변수로 인해 제한적인 해석을 지닌다. 그러나 동물생체정량과 비생체정량에서 얻어진 유용한 정보 [다양한 노출경로를 통해 다수의 종에서 폐, 피부, 위장부위에 종양율 유발: Bap의 궁극적인 대사체인 diol epoxide의 구조-활성(bay region-carcinogenic activity)상관관계: diol epoxide와 DNA와 반응하여 DNA 변성 및 염기쌍 분열 초래:diol epoxide는 미생물 및 포유동물세포에서 강한 직접활성 돌연변이원성]을 고려해 볼때 동물에서의 발암활성은 사람에서도 암이 발생할 수 있음을 예측한다. 따라서 BaP을 사람에 대한 유력한 발암물질(probable human ca

rcinogen)으로 분류한다.

2. 노출 평가: BaP는 대기중으로 배출되는 지점이나 근처에서 검댕이나 비산재와 같은 운반입자에 흡착된다. BaP이 흡착된 운반입자들은 대부분 2.1μm 이하의 미세입자로서 폐내 침착율을 더욱 증가시킨다. 이러한 BaP의 오염원은 도시대기의 경우 대부분 자동차 배출물질에 기인한다. 대기중으로 배출된 BaP은 환경중 오존 및 질소산화물과 반응하고 광화학적 반응을 일으켜 대기중 Bap의 이동 및 변경에 지대한 영향을 미친다. 또한 BaP은 지용성으로서 환경중에서 오랜 지속성을 나타낸다. 서울시 일부지역에서 이와 같은 BaP의

대기중 농도는 1.33∼4.64ng/m**3으로서 평생동안의 일일 노출량은 7.54∼24.30 ng/m**2/day으로 산출되었다.

3. 용량-반응 평가: 저용량에서의 위해도를 예측하기 위해 체표면적을 근거로 한 용량전환 및 수학적 모델을 적용한 결과, 실험용량 부위에서는 대체적으로 내성분포모델(tolerance distribution model : probit, logit, Weibull)이 관찰된 반응과 잘 일치하였으나 저용량에서는 예측이 불가능하였다. 따라서 저용량부위에서 보수적인 위해도를 나타내는 일점 모델 및 다단계 모델(one-hit and multistage model)을 채택하여 단위 위해도 추계

치 및 실제적인 안전용량을 예측한 결과, 일점모델에서는 각각 2.8 × 10**-7, 3.4ng/m**3이었고 다단계 모델에서는 각각 5.2 × 10**-7, 1.9ng/m**3으로서 다단계 모델이 더 보수적인(conservative) 위해도 추계치를 제공하였다.

4. 위해도 결정: 노출 평가와 용량-반응 평가를 조합한 결과, 서울시 일부지역의 현 오염도 수준에서, 일점모델의 경우 백만명당 0.37∼1.30명, 다단계 모델을 적용하였을 때는 백만명당 0.69-2.41명의 발암 확률을 나타내었다. 반면역학연구를 토대로 결정된 WHO의 비교 위험도 모델을 적용한 결과 십만명당 11.90∼41.76명의 발암확률을 나타내어 다단계 모델에 비해 약 173배 높은 결과를 나타내었다.

본 연구에서의 단위 위해도 추계치는 BaP의 단독 효과를 나타내는 반면 WHO와 단위위해도 추계치는 BaP의 단독효과를 나타내는 것이 아니라 BaP을 지표로 하여 다환 방향족 탄화수소류의 발암성 분획에 대한 복합효과를 나타낸 것이므로 이러한 결과의 차이가 나타

난 것 같다. 따라서 본 연구 결과 BaP의 단독물질에 대한 발암효과(백만명당 1명의 수준)를 고려해 볼 때는 실질적인 위해도는 없을 것으로 간주되나 대기중에는 실제적으로 BP을 포함하는 다환 방향족수소류(발암분획)가 동시에 공존하기 때문에 그 영향을 고려하여 WHO의 결과(십만명당 10명이상)를 수용할 때는 서울시 일부지역의 대기오염도는 심각한 것으로 해석된다.



[영문]This study is to set up methology of risk assessment as a basis for establishment environmental standard and a system of safety evalution of chemical(s). On the basis this methology, risk assessment was performed through the main four steps: hazard indentification, exposure assessment, dose-response assessment and risk characterization for benzo(a)pyrene in ambient air of Seoul. The results are summarized as follows:

1. HAZARD IDENTIFICATION: Benzo(a)pyrene is an animal carcinogen. On the other hand, evidences of carcinogenesis in human is limited. Taking account for the available information which obtained from bioassay and nonbioassay observation,

benzo(a)pyrene is positive carcinogenic activity at various exposure routes, in several species and at many target organs; diol epoxide, ultimates metabolite of benzo(a)pyrene has structure-activity(bay region-carcinogenic activity), reacts

DNA, and is a direct potent mutagen in microorganism and mammalian cells. The factors listed above can support for the predictability of the animal response for human. Therefore benzo(a)pyrene is classified a probable human carcinogen.

2. EXPOSURE ASSESSMENT: Benzo(a)pyrene is virtually adsorbed on carrier particles such as soot and fly ash at or near the point of emission into the ambient air. These carrier particles which are fine ones below 2.1μm, increase deposition rate of pulmonary region. In the case of metropolis including Seoul, the air pollution of benzo(a)pyrene was attributed to vehicles emission. Benzo(a)pyrene emitted into the ambient air reacts with other ambient pollutants such as ozone and nitrogen oxides and forms other products by photochemical reactions. These reactions

influence on persistence, transport and transformation of benzo(a)pyrene in ambient air. The concentration of benzo(a)pyrene was 1.33-4.64ng/m**3 and the average daily

lifetime exposure of one was calculated as 7.54-24.30ng/m**2/day.

3. DOSE-RESPONSE ASSESSMENT: Using dose scaring based on body surface area for extrapolation from animal to human, and mathematical models for extrapolation from high-dose to low-dose region, author estimated risk at low-dose region. The response probabilities estimated by tolerance distribution model(probit, logit, Weibull) were consistent with the observed ones at experimental dose region, but the unit risk estimates of these models were too low to be used. Therefore one-hit and multistage model were selected to provide conservative risk. As a result,

lifetime unit risk estimates and virtually safe dose were calculated. One-hit model provided 2.8 × 10**-7, 3.4ng/m**3 respectively and multistage model 5.2 × 10**-7, 1.9ng/m**3(more conservative) respectively.

4. RISK CHARACTERIZATION: Combining two step(exposure and dose-response assessment), risk was calculated. lifetime excess risk estimates was calculated as 0.37-1.30 persons/ million persons(one-hit) and 0.69-2.41 persons/million persons(multistage model). On the other hand, lifetime excess risk estimates was

calculated as 11.90-41.76 persons/ a hundred thousand persons in the case of relative risk model by World Health Organization which based on epidemiologic studies. Therefore lifetime excess risk estimates using relative risk model was about 173 times higher than one using multistage model.

The unit risk estimates of this study presents single effects of only benzo(a)pyrene, on the other hand, that of study of WHO presents not single effects of benzo(a)pyrene but combining effects of carcinogenic fractions of polynuclear aromatic hydrocarbons using BaP as a surrogate. Therefore, the difference between these results can be explained. Taking account for carcinogenic effects of only BaP, about a person per a million persons level(this study result) was considered as without virtual risk. But, because PAHs including BaP exsits simultaneously in ambient air, if the study result of WHO(above ten persons per a hundred thousand persons) were accepted, air pollution is a serious situation at some areas in Seoul.
