[한글]

연계에 존재하는 병원성 자유생활 아메바의 감염은 주로 비강후점막(嗅粘膜)을 통해 이

루어지며 후신경(嗅神經)을 따라 중추신경계에 침범하여 뇌의 기저부에 괴사성 출혈성 수

막뇌염을 일으킨다고 하였다(Martinez등, 1973). 비록 이 아메바의 병원성을 설명할만한

기전은 확실히 밝혀지지 않았지만 숙주세포를 빠르게 파괴시킬 수 있는 능력이 있음을 볼

때 수막뇌염을 일으키기까지 전과정에서 cytolytic process가 이루어질 것이라고 하였고

(Lowrey및 Mc Laughlin, 1985) 여러 동물세포에 비특이적 cytolysis현상이 일어난다고 보

고되었다(Marciano-Cabral등, 1982). 병원성 아메바의 cytolytic process가 주로 가수분

해 효소활성과 관계 깊다는 연구들이 70년대 후반부터 Entamoeba spp.와 Naegleria sap.

에서 보고되었지만 Acanthmoeba spp.에서는 가수분해 효소의 활성에 관한 연구는 찾아볼

수 없다.

이에 본 연구에서는 병원성이 없는 것으로 알려진 Acanthamoeba royeba를 대조로하여

병원성이 있는 Acanthamoeba culbertsoni의 가수분해 효소 활성도를 측정하고 이들을 비

교하고자 하였다. 즉 이 아메바를 마우스에 감염시켜 병원성이 있음을 재확인하였고, CGV

(Bactocasitone, Glucose, Vitamins)배지에서 배양할 때 시간에 따른 효소활성도를 측정

하여 비교하였으며, 또한 조직세포에 이들 아메바를 혼합하여 세포독성의 비교 및 처리

시간에 따른 효소활성도를 측정하여 다음과 같은 성적을 얻었다.

A. culbertsoni를 마우스에 감염시켰더니 15일내에 마우스 모두가 사망하였으며, 평균

생존기간은 8.5일이었고, 아메바성 수막뇌염으로 사망했음을 알 수 있었다. 반면에 A. ro

yreba를 감염시킨 마우스에서는 수막뇌염이 발생되지 않았다.

CGV배지에서 무균 배양하였을 때 아메바의 증식률을 보면 A. culbertsoni에 있어서 gen

eration time은 30.73±7.06시간으로 A. royreba에서의 73.37±13.34시간에 비해 약 2.5

배의 증식률을 나타내었다.

CGV배지에서 두 종(種)아메바를 배양하여 아메바 추출물 및 배양액에서 가수 분해 효소

의 활성을 측정한 결과 A.royreba에 비해 A.culbertsoni 세포의 추출물 및 배양액에서 현

저히 활성도가 높게 측정되는 가수분해 효소로는 acid phosphatase, β-N-acetyl galacto

saminidase, β-N-acetylglucosaminidase, α-mannosidase, neutral proteinase, acid pr

oteinase였다.

표적조직 CHO세포에 혼합한 A.culberhoni는 대조군에 비하여 강한 세포독성을 나타내었

으며 CHO세포에 혼합후 48시간 후에는 80%, 72시간후에는 95% 이상의 CHO세포가 사멸하는

것을 알 수 있었다. 반면에 A.royreba를 혼합하였을 때 별다른 세포독성을 나타내지 않

았다.

CHO세포에 혼합한 A.culbertsoni의 가수분해 효소 활성도를 경과 시간별로 측정하였다.

CGV배지에서 배양시간이 지남에 따라 아메바내보다 그 배양액에서 더 높은 효소 활성도

를 나타내었던 acid phosphatease, β-N-acetyl galactosaminidase, β-N-acetylglucosam

inidase, α-mannosidase, acid photeinase는 혼합 120시간 동안 아메바내에서 더높은 효

소활성도를 나타내었으며, neutral proteinase의 활성도는 경과시간이 지남에 따라 아메

바내에서는 일정하였지만 그 배양액인 EBSS용액내에서는 점점 높아짐을 알 수 있었다.

이상의 성적으로 보아 마우스에서 아메바성 수막뇌염을 일으키는 A.culbertsoni에서 여

러 종류의 가수분해 효소들의 활성도가 비병원성 A.royreba에서와 비교할 때 현저히 차이

가 있었음을 알 수 있었다. 또한 표적조직 CHO세포에 대한 A.culbertsoni의 세포독성이

강함을 알 수 있었고, 이때 몇 종류의 가수분해 효소들의 활성도가 높음을 측정할 수 있

었으며, 이러한 소견이 A.culbertsoni의 병원성과 관련이 있을 것으로 추측되는 바이다.

[영문]

Several species of free-living amoebae have been recognized as the causative

agents of human primary amoebic meningoencephalitis (PAME). These pathogenic

amoebae invade the central nervous system through olfactory mucosa mainly when the

victim has been swimming in the contaminated freshwater.

Specific or non-specific cytolytic processes of these amoebae causing

meningoencephalitis have been emphasized by many researchers even though their

pathogenesis has not been well known yet. A few reports on the cytolytic ability

relating to hydrolases in Entamoeba spp. and Naegleria spp. have also been

published since the latter half of 1970's. However, no information on hydrolase

activities in Acanthamoeba spp. has been available so far.

Hydrolases in Acanthamoeba culbertsoni which is regarded as a pathogenic species

of free-living amoebae were assayed as compared with those in a non-pathogenic

species, Acanthamoeba royeba. Pathogenicity of these two species was reconfirmed

through experimental infection by using BALB/C mice, and hydrolase activities and

cytotoxic effects between pathogenic and non-pathogenic species were compared in

the trophozoites cultured in CGV media and in CHO cell line, respectively. The

results are summarized as follows:

1. The mice infected with A.culbertsoni were all dead 15 days after nasal

inoculation, and the mean survival time was 8.5 days. Also the mice infected with

this pathogenic species manifested typical meningoencephalitis, whereas the mice

infected with A.royreba did not.

2. Hydrolases detected both in the cell extracts and culture media wee acid

phesphatase, β-N-acetyl galactosaminidase, β-N-acetyl glucosaminidase,

α-mannosidase, neutral proteinase and acid proteinase, all of which were detected

with remarkably higher rate in A.culbertsoni than in A.royreba.

3. A.culbertsoni revealed strong cytotoxic effects for the target CHO cells,

whereas A.royeba did not show any specific cytotoxic effect for the target cell

line. About 80% of the target cells mixed with A.culbertsoni were dead 48 hours

after cultivation, and more than 95% of the target cells were dead 72 hours after

cultivation.

4. Hydrolase activities in A.culbertsoni cultured with the target cell line were

assayed according to the culture time. The activities of acid phosphatase,

β-N-acetyl galactosaminidase, β-N-acetyl glucosaminidase, α-mannosidase and acid

proteinase in this pathogenic amoeba were detected higher in amoeba extracts than

in culture media up to 120 hours after cultivation, but after 120 hours of

cultivation these activities were detected higher in culture media than in the

amoeba lysates. Neutral proteinase activity in A.culbertsoni increased gradually

more in EBSS medium than in the lysate specimens although the activity in the

extracts was generally steady according to the cultivation time.

In summarizing the above results, it is concluded that there was difference of

hydrolases activities between pathogenic A.culbertsoni and non-pathogenic A.

royreba, and that some hydrolase activities were detected remarkably higher in A.

culbertsoni which revealed strong cytotoxic effect to the target CHO cell line than

in non-pathogenic A.royeba.
