Abstract
[한글]

세포매개 임파구용해반응 (cell mediated lympholysis; CML)에 있어서 세포독성임파구(

cytotoxic lymphocyte; CTL)의 표적인자(target determinants)로서 HLA-A 및 B항원과 HLA

-C항원 등이 중요한 역할을 하는 것으로 보고 되어 있다. 한편 최근에는 단핵구, Epstein

-Barr virus에 의하여 형질전환(transformation)된 B임파구 및 pokeweed mitogen처리 B

임파아세포 등을 표적세포로 이용한 세포매개 임파구용해시험에서 HLA-DR 항원 역시 세포

독성임파구의 표적인자로 작용함을 보고하였다. 그러나 동일한 HLA항원을 가진 형제간에

는 세포독성을 관찰할 수 없었으나 동일한 HLA항원을 가진 비친족간에는 세포독성을 관찰

할 수 있으며 세포매개 임파구용해반응에서 표적세포로 사용한 임파구가 자극임파구와 동

일한 HLA-A, B 및 C항원을 가지고 있지 않으나 CML반응에서 양성을 나타내며 표적임파구

와 자극임파구가 동일한 HLA항원을 가지고 있으나 CML반응에서 음성반응을 관찰할 수 있

고, HLA항원 자체는 아니나 HLA complex의 지배를 받으며 두개의 일배체형 (haplotype)의

보완작용 (complementation) 효과를 보이는 새로운 표적인자를 관찰할 수 있다는 보고

등으로 보아 세포매개 임파구용해반응 과정에서 혈청학적으로 규명할 수있는 HLA-A, B, C

및 DR항원 이외에 다른 표적인자의 존재를 암시하여 주고 있다.

이에 저자는 세포매개 임파구용해반응에 관여하는 표적인자로서 HLA-A, B 및 DR항원의

상대적 기여도를 측정관찰하고 새로운 CML 표적인자를 규명하기 위하여 본실험을 실시한

바 강한 연관불균형을 나타내는 A^^3:B^^7 항원 으로 감작된 CTL에 의하여;

1. CML 표적세포의 HLA-B^^7 항원은 HLA-A^^3 항원과 공존여부에 관계없이 단독으로도

강한 CML 표적인자로 작용하였다.

2. HLA-A^^3 항원은 CML 반응에서 E△ 3. 7. 2에 의하여는 약한 표적인자로 작용하였으

나 E△ 3. 7. 0와 E△ 3. 7. 5에 의하여는 비교적 강한 표적인자로 작용하였다. 이는 CTL

유도를 위한 감각과정에서 반응임파구가 low responder와 high responder에 의한 차이의

가능성과 DR^^2 항원자체 혹은 DR^^2 유전자좌와 연관되어 다른 유전자가 A^^3 항원을 인

지하는 반응에 영향을 줄 수 있는 가능성을 암시하여 준다.

3. 혈청학적 방법으로 확인된 HLA-B^^7 항원을 가지고 있는 표적임파구중 JN(Al, 29; B

5, 7; Cw-; DR^^2) 표적임파구가 B^^7을 가지고 있는 다른 표적 임파구 FP와 다른 CML 반

응을 보이는 것은 현재 사용되는 혈청학적 방법으로는 인지할 수 없는 JN임파구에만 표현

되는 B^^7의 변이 인자의 존재 가능성을 암시하여 준다.

4. PHA활성 표적임파구를 이용한 CML반응 실험에서는 DR항원의 표적인자로서 기여는 관

찰할 수 없었다.

5. 감작임파구와 동일한 HLA항원을 가지고 있지 않으나 높은 CML 반응을 보이는 수종의

문외임파구(outliers)를 관찰할 수 있는 것으로 보아 여러개의 새로운 CML표적인자를 지

배하는 유전자좌의 존재 혹은 여러개의 대립인자를 가지고 있는 단일 유전자좌의 존재를

암시하여 주는 것으로 생각한다.

Earlier studies on the genetic control of cell lmediated lympholysis (CML)

indicated that serologically defined HLA-A and B alloantigens(Bonnard et al., 1973:

Eijsvoogel et al., 1973; Miggiano et al., 1972; Trincheri et al., 1972) and some

HLA-C antigens(Grunnet et al., 1976; Kristensen and Grunnet, 1975) can serve as

target determinants with varying strength. Recently, it has been shown that the

HLA-DR antigens can also serve as CML target determinants by using monocytes

(Feighery and Stastny, 1979) or Epstein-Barr virus transformed

B-lymphocytes(Albrechtsen et al., 1979). It was also shown that it was possible to

demonstrate target determinants exclusively expressed on pokeweed mitogen

stimulated B lymphoblasts (Johnsen, 1980).

There is, however, considerable evidences that determinants other than HLA

antigens can be recognized by cytotoxic T lymphocytes(CTL). The existence of target

determinants distinct from HLA-A, B and C antigens has been shown by studies of a

number of investigators. There by now are a number of indications that determinants

other than HLA antigens function as target determinants in CML. Such evidences come

firstly from observations of CML in combinations between HLA-A, B, C identical,

unrelated individuals(Goulmy et al., 1975: Grunnet and Kristensen, 1975; Schapira

and Jeannet, 1974) in contrast the negative findings with HLA-identical siblings

(Sondel and Bach, 1976). Secondly, combinations have been identified giving

reproducible patterns of cytolysis which could not be explained by HLA-A, B, C

antigens sharing between stimulating and target lymphocytes(Kristensen et al.,

1974; Mawas et al., 1974) conversely combinations have been identified yielding no

cytolysis despited well-defined HLA-antigenic identity between stimulating and

target lymphocytes(Kristensen et al, 1975; Willumsen and Heron, 1974). A third

piece of evidence comes from the observation of target determinants other than HLA

antigens but its recognition may be restricted by the HLA complex(Schendel et al.,

1978).

In an attempt to study the relative strength of the HLA-A, B, and DR antigens as

target determinants in CML and identify new CML target determinants, the following

experimental model has been established. Firstly, CTL have been generated against a

single HLA haplotype using haploidentical first degree relatives. Secondly, large

number of CTL have been generated from a single priming and these have been used

for all experiments, thereby overcoming potential variability with each priming.

Thirdly, cells have been primed against an HLA haplotype which displays strong

linkage disequilibrium. Fourthly, these effector cells have been used to assess the

relative strength of HLA antigens as target determinants using target cells bearing

various combinations of the sensitizing HLA antigens. Conceivably, other HLA-linked

CML target determinants might also display strong linkage disequilibrium with these

haplotypes. Such determinants could potentially be detected in the CML assay in

spite of lack of detection by present serology.

Using this model, five sets of cytotoxic effector cells were generated in five

different families against the A3:B7 serological determinants combined with

different DR antigens. When tested against a panel of cells bearing combinations of

the HLA-A, B and DR antigens it was shown that the HLA-B7 antigen was as strong CML

target determinant alone as it was in the presence of HLA-A3. And one particular

target lymphocyte JN(Al, 29; B5, 7; Cw-; DR2) bearing HLA-B7 behave differently

from other HLA-B7 bearing target lymphocytes. This suggests the possibility that a

determinant of B7(variant of B7) is expressed on cell JN, but which cannot be

detected serologically.

In terms of the contribution of HLA-DR antigen as a CML target determinants, our

results suggest that HLA-DR antigens are not strong determinants in CML. The

strength of the HLA-A3 antigen varied. With effector cells primed to the A3:B7:DR2

haplotype, the A3 antigen alone behaved as a weak target determinant. When the same

target cells were tested with the other three sets of effectors, the A3 antigen

behaved as a strong target determinant. The possibility can be raised that either

low and high responders to A3 have been detected or the presence of the DR2 locus

or genes linked to it is influencing the ability to recognize and respond to A3.

A number of target cells("outlilrs") lacking the stimulating antigens were

identified as being killed by specific effector cells. These data suggest either a

number of new CML target determinants or multiple allelea of a single new locus

exist
